Effect Of Taohe Chengqi Decoction On CTGF Expression In Renal Tissue Of Rats With Chronic Renal Failure

Objective: Through observing the effects of Taohe chengqi decoction on blood routine,renal function,renal pathological morphology and CTGF in the signaling pathway of TGF-?/smad of rats with chronic renal failure,the mechanism of action for this method that can alleviate or delay chronic renal failure was explored,and then enriches treatment ideas of chronic renal failure,thus providing the experimental testimony to clinical treatment of chronic renal failure.Methods: 125 Wistar rats were divided into 5 groups.Group A: blank group,as the health group of this experiment to be fed,did not do any treatment.Group B: sham operation group,intending to simulate surgical trauma,observed whether the model surgery(only the bilateral kidneys of the model are removed by membrane)had an impact on its experimental model indicators.Group C: model group,only model surgery,without any drug treatment,was intended to observe the progress of chronic renal failure of the model without any drug intervention.Group D: Traditional Taohe chengqi decoction group,administering Taohe chengqi decoction to the model by gavage after the mold operation,was aimed to observe the effect of on its model chronic renal failure.Group E: NDQ group,injecting the model with NDQ granule by gavage after mold making surgery,was designed to observe the effect of NDQ on the model chronic renal failure,and was compared with the experimental data of Chinese medicine group.Model production: After one week of adaptively feeding experimental animals,5/6nephrectomy was performed on rats in the model group,the traditional Chinese medicine group and the NDQ group to replicate the CRF model.2/3 left lateral nephrectomy was performed on those three groups of rats under chloral hydrate anesthesia,and the total right lateral nephrectomy was performed after one week of operation while renal membrane decollement of bilateral kidney was performed in the sham surgery group.Within three days after surgery,penicillin was injected daily into the abdominal cavity to reduce the possibility of infection.All experimental animals were fed according to blank group after four weeks of building models,the fake operation group with normal saline,the traditional Chinese medicine group with Taohe chengqidecoction,the NDQ group with NDQ granule and the model group with normal saline.Besides,in the third week after the mold was made,blood was collected from the fundus venous plexus of the model to detect RBC,Hb,Scr,and BUN to testify whether a successful model was made.After eight weeks of continuous lavage,blood was collected from the Aorta abdominalis under chloral hydrate anesthesia to detect RBC,Hb,Scr,BUN.The left kidney tissue was taken and divided into two parts.One part of kidney tissue placed in Cryogenic Vials to use Western Blot to detect the protein content of the left kidney tissue CTGF,Smad 3,and Smad7 and use Real-Time PCR to detect the expression of CTGF,Smad 3,and Smad7 while the other part,stored in paraformaldehyde,was use to observe the pathological conditions of the kidney tissues of each group with light microscope after treatment,and then pictures were collected to compare the change of each group.Results:(1)Compared with the model group,the content of RBC and Hb in the Tao he Chengqi Decoction group had varying degrees increased.(P<0.05).(2)The results t hat serum SCr and BUN of Taohe Chengqi Decoction group were lower than those ofmodel group and uremic clearance group(P<0.05)have statistically significance.(3)Un der light microscope,HE staining of renal tissue showed that left renal tissue structure in the blank group and the sham operation group were normal.In the model group,the left renal tissue structure was disordered,glomerular edema and hyperplasia of ren al interstitial fibers,and inflammatory cell infiltration was observed.Other than that,co mpared with uremic clearance group and model group,the degree of renal fibrosis wa s obviously improved in Taohe Chengqi Decoction.(4)the results of Western Blot sho wed that the protein expression of Smad3,CTFG in the left kidney of the Taohe Chen gqi decoction group was lower than that of the model group(P<0.05).The expression level of Smad7 protein was up-regulated(P<0.05).(5).Real-time fluorescence quantitat ive qPCR showed that the expression of Smad3,CTFG mRNA was down-regulated(P<0.05)and the expression of Smad7 mRNA was up-regulated(P<0.05)compared with t he model group.Conclusion:1.Taohe chengqi decoction can improve the general situation of rats with 5/6nephrectomy and,to a certain extent,can alleviate the symptoms of renal failure,treat anemia,slow renal function degradation,and reduce Scrand and Bun.2.Through Taohe chengqi decoction inhibited the expression of Smad3 and CTGF by affecting the TGF-? / Smad signal path and promoted the expression of Smad7,it can be learned that Taohe chengqi decoction,to a certain degree,can prevent the deterioration of renal units and delay the progress of renal fibrosis.
