Objective: The increasing severity of overweight and obesity have become a major social problem in the world.Obesity can cause a series of chronic metabolic diseases,and it does significant harm to human society development and the health of residents.The actual oil intake of urban and rural residents(42.1g/d)exceeds one third of the World Health Organization's recommendations in China.In this study,based on the traditional dietary habits in which Chinese residents mix vegetable oils and animal oils,mice were used as experimental animal models to study the effect of actual oil intake(lard oil,sunflower oil,and compound oils)on the body's anti-obesity,lipid metabolism and blood glucose homeostasis in China's urban and rural areas.Provide reference for the reasonable daily use of oil for our residents to reduce overweight and obesity,and it provides a reference of the reasonable choice daily dietary oils to our residents,and reduces the prevalence of overweight and obesity.Methods: Thirty male C57BL/6J mice of 6w age were selected and the initial body weight was about 23 g.They were randomly divided into 3 groups,lard group(LO group),sunflower seed oil group(SO group),lard blend sunflower seed oil group(BO group)respectively.According to the actual average oil intake of urban and rural residents in China(42.1g/d),the feed oil level of mice was 6.3%.Lard oil,sunflower oil,and lard blend sunflower seed oil were added in feed separately.All mice were adaptive fed in one week before the experiment,according to different experimental groups fed the corresponding feed.The experiment period was twelve weeks.Anesthesia was performed and blood was collected from the eyeballs.Serum,liver,and adipose tissue were collected.Body fat,blood lipids,hepatic lipids,hepatic lipid deposition,hepatic antioxidant capacity,and indicators related to glucose metabolism were measured.Results: 1)The effect of different of oils on body fat deposition in mice.The final body weight and body weight rate of SO group were higher than the LO group (P<0.05),but those were significantly higher than the BO group(P<0.01).The SO group had higher fat weight and fat cell cross-sectional area than the LO and BO groups(P<0.05).The concentration of fat TC in the SO group increased compared with the LO group(P<0.05),and markedly increased compared with the BO group(P<0.01).2)The effect of different oils on blood lipids and the metabolism of blood lipids in mice.The BO group had lower the concentrations of serum TG,LDL-C and FFA than the SO group(P<0.05),and those were significantly lower than the LO group(P<0.01).The level of serum ADPN in the BO group was higher than the SO group(P<0.05),and it significantly higher than the LO group(P<0.01).The level of serum Leptin in the LO group increased compared with the SO and BO groups(P<0.05).3)The effect of different oils on liver fat and liver lipid metabolism in mice.The concentration of liver TG and liver oil red staining in the SO group were higher than the LO and BO group(P<0.05).The SO group had significantly higher the activity of FAS in liver than the LO and BO groups(P<0.01).The activity of liver HSL in the SO group was lower than the BO group(P<0.05).4)The effects of different oils on liver function and liver antioxidant ability of mice.The activity of liver AST in the LO group was significant diminished in comparison with the SO and LO groups(P<0.01),and the activity of liver ALT in the LO group was lower than the SO group(P<0.05).Compared with the BO group,the level of liver T-AOC was decreased in the SO and LO group(P<0.05).The BO group had higher the activity of liver SOD than the SO group(P<0.05),and it had significantly higher than the LO group(P<0.01).5)The effect of different oils on the regulation of blood glucose in mice.The level of Blood glucose in the LO group was lower than the SO and BO groups(P<0.05),and the BO group had significantly lower than the SO group(P<0.01).The level of serum insulin in the LO group was increased compared with the SO and BO groups(P<0.01).The LO group had higher the level of serum GLP-1than the BO group(P<0.05),and it was markedly ascended compared with the SO group(P<0.01).The SO group had higher the level of serum resistin than the LO and BO groups(P<0.05).Conclusions: 1)Compared with sunflower seed oil,the intake of lard oil and lard blend sunflower seed oil decreased body weight,fat weight,body fat rate,adipocyte lipid content and adipocyte cross-sectional area,and reduces body fat deposition.2)Compared with lard oil and sunflower seed oil,the intake of lard blend sunflower seed oil could decrease the levels of serum TG and FFA,and it could reduce the levels of serum TC and LDL-C and improve blood lipids.3)The intake of lard blend sunflower seed oil could diminish the activity of FAS in the liver,increase the activity of HSL and the level of circulating ADPN,ultimately reduce the effect of lipid deposition and body fat deposition in the liver.It had a certain preventive effect on the occurrence of non-alcoholic fatty liver and obesity.4)Compared with lard oil and sunflower seed oil,the intake of lard blend sunflower seed oil could increase SOD activity and T-AOC level in the liver,thereby enhancing the liver's ability to resist oxidative stress.5)Compared with sunflower seed oil,proper intake of lard oil could decrease blood glucose by increasing circulating insulin levels.Lard blend sunflower seed oil could regulate blood glucose homeostasis by reducing serum resistin levels and increasing insulin sensitivity,and it had certain positive effect on preventing type II diabetes.
