Objective:Observation of Huayu Ruangan Capsule on Sugar in Rats with Non-alcoholic Fatty Liver Disease of Qi Stagnation and Blood Stasis Type?Lipid metabolism related of Superoxide dismutase?Malondialdehyde?Glutathione peroxidase?Glucokinase?Glucose6-phosphate?Stearic acyl coenzyme A-1 influence,Preliminary study on its mechanism of action,To provide experimental basis for clinical application of Huayu Ruangan Capsule in the treatment of non-alcoholic fatty liver.Method: 60 SD rats One week after normal adaptive feeding,randomized grouping was used,They were divided into normal control group and model group.Except for the normal rats,the model group was fed with self-made high-fat diet(15%lard+10% custard powder+70% Basic feed+5% White granulated sugar)induced diet,Establishment of a rat model of non-alcoholic fatty liver disease with qi stagnation and blood stasis by tail-clamping stimulation and reagent intervention.The model group was induced to feed on a high-fat diet for 8 weeks,Fifth week after the model was established,rats were clamped with long sponge oval forceps at 1/3 of the distal end of tail every day,in order not to damage the epidermis,so that the rats became angry,and then tore and wrestled with other rats,thus making the rats in safe cage irritable.Once a day,each stimulation lasted for 30 minutes.Under continuous stimulation for 30 minutes,the degree of fighting gradually intensified.Some rats may be scratched or bitten.To avoid inflammation interference,0.5% iodophor can be applied to the injured area to prevent infection and inflammation interference,and calf serum albumin is injected into the tail vein of the rats at regular intervals on Monday,Wednesday,week 5 and Wednesday 6 of the model establishment.The concentration was 50g/L and the dosage was2.5ml/Kg,Until increased to 1.0 ml/kg per day and continued until the end of modeling.Feed every weekend and every other day,weigh their weight,24 hours of diet and drinking water.The hair color,tongue color,eye color,skin color and irritability of rats were observed during modeling.After 8 weeks,3 rats in the model group and 3 rats in the normal group were randomly selected,and femoral artery blood was taken after execution,Activities of superoxide dismutase,malondialdehyde,glutathione peroxidase,glucosylase,6-phosphate glucose and stearyl coenzyme A1 were analyzed by kit.1 g fresh liver tissue was weighedand sliced.The morphological changes of liver tissue were observed under light microscope.Result:1.Compared with the control group,the liver volume of the model group was signi ficantly increased compared with the control group.The liver wet weight ratio of the model group was greater than that of the control group.Compared with the control gr oup,the rats in the model group were dull and emotionally irritable.After drug interv ention,the color and mood of the rats in the drug intervention group were superior to those in the negative control group.2.The overall data of rat liver wet weight showed that the model group was higher than other groups,and the liver tissue of the rats was analyzed.The results showed that the liver index of the model group rats(yin group)was higher than other treatme nt groups.In the middle,the high dose of traditional Chinese medicine and the rosuv astatin treatment group(positive group)were closer to the liver index.3.The liver tissues of the experimental rats were observed under light microscope.The structure of the control group was almost complete and clear.The model group s howed diffuse hepatic steatosis.The degree of steatosis of diffuse liver cells in each t reatment group was lighter than that in the model group.4.The results showed that the MDA activity value of the model group was slightly different from that of the normal group and the treatment group.There was significant difference in MDA between the treatment group and the model group.The SOD and GSH-PX activity model groups were significantly different from the normal group.Th ere was significant difference between the group and the remistatin group(P<0.05).5.Compared with the control group,the content of the model group was reduced to a certain extent,and the difference between the levels of the treatment group and the model group was small.The reason for the analysis was attributed to the stress respo nse and serum storage time at the time of blood collection.6.Compared with the normal group,the expression of GCK in the model group,the middle dose group and the high dose group was significantly lower,while that in the rosuvastatin group and the low dose group was slightly lower.Compared with the normalgroup,the expression of G-6-P mRNA in the low-dose group and the high-dose group was significantly higher(P < 0.05).Conclusion:1.The method of legging stimulation + high-fat-induced diet + injection of norep inephrine + injection of calf albumin can successfully produce an animal model of qi stagnation and blood stasis non-alcoholic fatty liver.2.Huayu Ruangan Capsule has certain therapeutic effects on qi stagnation and bl ood stasis type non-alcoholic fatty liver,especially for the damage caused by oxidative stress and lipid peroxidation in nonalcoholic fatty liver.3.For non-alcoholic fatty liver patients with diabetes(islet dysfunction),Huayu Ruangan Capsule has certain effects on improving glucose and lipid metabolism in the body,regulating glucokinase and glucose 6 phosphatase in the body,and enhancing m etabolism in the body.
