This paper mainly chose the roots of the poisonous Tripterygium hypoglaucum(Levl.)Hutch of Tripterygium from Leishan County,Guizhou province as the research object.The extraction fractions of ethanol from the roots of Tripterygium hypoglaucum(Lévl.)Hutchins.in Guizhou province were separated and purified by normal phase column chromatography,normal phase pressurized column chromatography,positive pressure column chromatography,Sephadex LH-20 gel column chromatography,reversed-phase column chromatography,semi-preparative HPLC and recrystallization.Their structures were identified by UV?EI-MS?ESI-MS?NMR and combined with physical deconstants termination and control of thin layer chromatography(TLC).18 compounds were isolated and identified from the ethanol extract of Tripterygium hypoglaucum(Levl.)Hutch in Guizhou province.They are3-O-Acetyloleanolicacetic anhydride(1),Triptonoterpene(2),3-Oxo-olean-12-en-28-oic acid(3),?-Sitosterol(4),Friedelin(5),?-Sitosteryl palmitate(6),Celastrol(7),Emodin(8),Wilforlide A(9),Triptoquinone B(10),Ent-kauran-16?,19-diol(11),Quinone 21(12),?-Amyrin(13),?-Amyrin acetate(14),Triptolide(15),Triptophenolide(16),Oleanolic acid(17),?-Daucosterol(18).Compounds 6 and 11 were isolated from this plant for the first time,and meanwhile compound 11 was also isolated from the genus of Tripterygium for the first time.The antimicrobial activities of 18 compounds were screened by 96-well plate microdilution method.Compounds 3,7,8 and 10 were found to have antimicrobial activities against on Staphylococcus aureus,Pseudomonas aeruginosa and Ralstonia solanacearum.The MIC values of compounds 3,7 and 8 ranged from 2~16 ?g/mL.Compounds 10 and 12 had bacteriostatic effects on Ralstonia solanacearum,compounds 7 and 12 had bacteriostatic effects on Agrobacterium tumefaciens,and compounds 9,10,12 and 16 had bacteriostatic effects on Erwinia carotovora subsp.carotovora.Furthermore,it was found firstly that compounds 3 and 7 had obvious bacteriostatic effects on Pseudomonas aeruginosa and Rastonia solanacearum.In this paper,the extraction,seperation and bioactivity of Tripterygium hypoglaucum(Lévl.)Hutchins.from Guizhou province were systematically studied,which provided the experimental support and new research strategy for further study on Tripterygium hypoglaucum(Lévl.)Hutchins.
