| Objective: To study and explore the role of autophagy-related proteins P62 and ULK1 in the pathogenesis of polymyositis(PM)/dermatomyositis(DM).Methods: 1.The hematoxylin-eosin staining method was used to observe the microscopic changes of muscle tissue in PM/DM patients and control group.2.Immunohistochemical technique was used to detect the levels of P62 and ULK1 in muscle tissues of PM/DM patients and healthy controls.The data were statistically analyzed using SPSS 19.0 statistical software.Result:1.HE showed that the PM group and the DM group had muscle tissue atrophy,muscle fiber rupture and obvious inflammatory cell infiltration compared with the control group.The inflammatory cells infiltrated in the DM group were more obvious than the PM group.2.The expression of P62 in the control group was higher than that in the PM group and the DM group(P62 in the PM group and DM group were 30%).%,significantly lower than the positive rate of the control group(85%),P <0.05),the difference was statistically significant;The positive expression of ULK1 in the muscle tissue of patients with PM and DM group was significantly higher than that of the control group(the positive rate of ULK1 in the muscle tissue of PM group and DM group was 70% and 60%,respectively,which was significantly higher than that of the control group).The positive rate of ULK1(10%),P<0.05),the difference was statistically significant,there was no significant difference in the expression levels of ULK1 and P62 in the muscle tissue between the PM and DM groups.3.muscle groups of PM group and DM group The expression level of P62 was negatively correlated with the total visual analog score of myositis activity evaluation(correlation coefficient r was-0.869,-0,813,P<0.05);the expression levels of ULK1 in muscle tissues of PM group and DM group were positively correlated with the total scores of myositis activity evaluation visual analogs(correlation coefficient r were 0.901,0,873,respectively,P<0.05));Conclusion: 1.It is suggested that P62 and ULK1 may participate in the pathogenesis of PM/DM through autophagy.2.The level of P62 protein decreases with the increase of disease activity,and the level of ULK1 protein increases with the increase of disease activity,.P62 and ULK1 may become new indicators for evaluating PM/DM activity. |
