CYNOMORII HERBA is a commonly used Mongolian herbal medicine,which is made from dried fleshy stems.It has the functions of enriching blood,tonifying kidney-yang,moistening intestines and relieving constipation.Cynomorium songaricum is rich in polysaccharides and other substances.Studies have shown that Cynomorium songaricum polysaccharide(CSP)has anti-aging and anti-oxidation effects but little research has been done on its immune activity.B cells are one of the most important immune cells in living organisms.They play an important role in the humoral immune components of adaptive immune system by secreting antibodies.In this study,Cynomorium songaricum was used as experimental material to obtain CSP samples(CSP1-CSP5)by enzymatic hydrolysis for different time(1-5 h).Based on the small test conditions,combined with chemical analysis,infrared spectroscopy and scanning electron microscopy,the pilot-study extract of CSP was extracted.The preliminary study of the process provides a theoretical basis for the pilot production of CSP.Next,mouse spleen lymphocytes were treated with different concentrations of the obtained samples(CSP1-CSP5)to examine their effects on proliferation ofprogenitor B cells(Pro-B),pre-B cells(Pre-B),and immature B cells(immature B).The results are as follows:(1)CSP obtained at different enzymatic hydrolysis time can promote the proliferation of Pro-B cell,Pre-B cell and immature B cells.The effect of CSP5 on B cells is particularly obvious,in which the number of Pro-B cells increased by0.14%~0.29%,immature B cells increased by 0.05%~0.08%,while the number of Pre-B cells decreased by 0.03%~0.36% under low concentration treatment,which may be that low concentration can promote the development of Pre-B cells to immature B cells.(2)With the increase of enzymatic hydrolysis time,the polysaccharide content of Cynomorium songaricum increased.Compared with CSP1,the polysaccharide content of the other four samples increased by 10.77%,19.88%,26.17% and 32.38%,respectively.After preliminary separation,the polysaccharide content of the water-elution part increased compared with that of the crude polysaccharide.(3)The results of pilot-scale production of CSP:enzymatic hydrolysis for 10 hours,extraction for 3 times,extraction for 3 hours and enzyme dosage for 5%.The yield of CSP was 17.18%~20.26%,polysaccharide content was 78.54%~89.76%,protein content was 1.22%~1.8%.After scanning by electron microscopy,it was found that the raw materials were damaged more seriously.
