Cynomorium songaricum Rupr.)belongs to Suoyang genus of Suoyang family,which is mostly distributed in Qinghai and Ningxia.It is often used to improve kidney yang deficiency,waist and knee impotence,etc.,with the effect of tonifying kidney yang,moistening bowels and so on.In this study,the antioxidant components of Suoyang were selected as the research goal.Suoyang collected from Chaidamu in Qinghai was crushed and extracted with 95% ethanol,then it was preliminarily separated by macroporous adsorption resin chromatography.and the antioxidant activity of each component was verified.The active components were separated and prepared by high performance liquid chromatography(HPLC)and semi-preparative high performance liquid chromatography(HPLC).Two active components were obtained,and the chemical structures of 9 monomer compounds were determined.the potential antioxidation mechanism of the effective components of Suoyang was discussed by the method of network pharmacology.Through the hierarchical virtual screening strategy of molecular docking-binding free energy calculation,15 active components with good binding to antioxidant-related proteins were excavated.The main results of this paper are as follows:The main results are as follows:The main results are as follows: 1.Six components were isolated and purified by macroporous resin.After the verification of DPPH antioxidant activity,two components with better activity were identified,which were 60% ethanol fraction and 80% ethanol fraction.The liquid phase analysis of two active parts obtained by macroporous adsorption resin was carried out by high performance liquid chromatography(HPLC).According to the analysis results,nine compound monomers were further separated and purified by semi-preparative chromatography,and the purity was more than 90%.The structures of the nine compounds were identified as ursolic acid,catechin,Gallic acid,rutin,quercetin-3-O-b-D-glucuronopyranosyl-4’-O-b-D-glucopyranoside,quercetin-3-O-b-D-glucuronide,Syriacatin,quercetin 3muro-(2’’-O-galloyl)-b-Dglucuronopyranoside,quercetin-3-O-b-D-glucuronopyranoside methyl,respectively.2.Using TCMSP database and disease gene database,35 antioxidant active components and 499 antioxidation-related disease targets were searched and screened.After intersection,34 core targets of antioxidant-related diseases were screened,and the Suoyang active component-target network was constructed.Through PPI data analysis,GO and KEGG enrichment of overlapping genes,the top 10 antioxidant activity targets,chemical components with good activity and related signal pathways were obtained.3.By fitting the antioxidation-related active protein targets with the chemical components of Suoyang by molecular docking technology,four active components with good binding to each target were screened,and their binding sites and binding modes were analyzed.the material basis of antioxidant activity of Suoyang was further clarified.This study provides data support for the utilization of plant resources of Cyclocyang,and further studies on antioxidant chemical components of Cyclocyang can be deepened in the future.Lead drug design and pharmacophore screening can be conducted for the effective chemical components screened,providing research basis for the comprehensive utilization of Cyclocyang in various aspects.
