Objective:Phthalic acid-(diethyl hexyl)esters have been used as plasticizers to increase product flexibility and durability and in our daily life.DEHP is a kind of known environmental endocrine disruptors,which can affect the normal endocrine activities of humans and animals and cause potential adverse effects on human health.In recent years,DEHP is used in large volumes extremely,DEHP is constantly released into the environment thro?g h various ways and ingested by the human body thro?g h many channels.A large number of epidemiological investigations show that DEHP has become one of the most common pollutants in the world and people are exposed to DEHP widely.On the basis of optimizing the DEHP metabolites detection method,we detect the concentratins of 2 DEHP metabolites in urine of pregnant women and evaluated the levels of pregnant women in shenyang,China.Combining the date of questiomaire survey and urine measurement results,preliminarily analyzing the possible factors influencing the levels of urine DEHP metabolites.Otherwise,combining with the clinical indicators of pregnant women examination,discussing the influence of DEHP exposure on the health of pregnant women in Shenyang,China,provide scientific basis for preventing the health harm of DEHP to pregnant women.Methods:In this study,we selected 850 pregnant who volunteered to participate in this study from shengjing hospital affiliated to China medical university in Shenyang,excluded occupational exposure history and disease history.In this study,we excluded 19 cases who failed to fill in the questionnaire and 37 cases who were not collected urine samples.Finally,we included 794 particpators into this study.urine samples of evey participants were collected within one week before or after delivery and detected the levels of 2 DEHP metabolites of the urine samples,MEHP and MEHHP,in gas chromatography tandem mass spectrometry:All the subjects were asked to complete a questionnaire,mainly including general demographic characteristics,nutritional supplements during pregnancy,environmental exposure,and the clinical test data of each subjects were collected at the same time.The epidemiological results and measurement data were evaluated by multiple linear regression of SPSS 20.0.Results:(1)This study optimized the gas chromatography tandem mass spectrometry method for determing of 2 DEHP metabolites simultaneously.The determination results showed that the regression coefficients of 2 DEHP metabolites were r>0.99 compared to the theory,and the recovery limits were MEHP 0.04?g/L,MEHHP0.02?g/L,the detectable rates are MEHP 98.75%,MEHHP97.36%,and the geometric means of 2 DEHP metabolites were MEHP4.62?g/L,MEHHP5.11?g/L.(2)Analyzing the possible influencing factors on pregnant women exposure to DEHP showed that house decoration P<0.05(?=0.133),distance between the house and the traffic trunk P<0.05(?=0.125),living in the city p<0.05(?=0.095),moldy house walls P<0.05(?=0.184)may be the risk factors,while the use of humidifier maybe the protection factor of exposure to MEHP P<0.05(?=0.134).(3)Analyzing the data of 2 DEHP metabolites and maternal clinical indicators s?g ested that the level of MEHP was correlation with the level of serum neutrophil percentage P<0.05(?=0.016),lymphocyte count P<0.05(?=0.017),cystatin C P<0.05(?=0.025),and urea P<0.05(?=0.024).Conclusions:.l.This study has successfully optimized GC-MS to detect the level of 2 DEHP metabolites in maternal urine.We find that DEHP can be determined in most urine samples,indicated that pregnant women exposure to DEHP in Shenyang,China.2.Analyzing the possible influencing factors on pregnant women exposure to DEHP showed that house decoration,distance between the house and the traffic trunk,living in the city,moldy house walls may be increase the exposure and the use of humidifier maybe reduce the expose.3.Analyzing the data of 2 DEHP metabolites and maternal clinical indicators s?gested that the level of MEHP was possibable influencing factor to increase the level of serum neutrophil percentage,lymphocyte count,cystatin C,and urea.
