The Taxus family,as one of the 56 endangered and rare medicinal plants in the world,is rich in chemical constituents,among which,taxane diterpenoids are characteristic components and the main active substances.It is found that taxane diterpenoids has anti-tumor,antiinflammatory,analgesic,anti-bacterial,anti-convulsion,anti-platelet aggregation,antiAlzheimer’s disease and other pharmacological effects.In 2017,foreign scholars found that taxane diterpenoids also have excellent hypoglycemic activity.Taxus baccata L.,a member of this genus,widely distributed in European countries,is one of the major sources of taxol’s raw materials.In view of the research gaps of the plant at home and abroad in the past five years and the shortage of relevant reports on domestic cultivars,the chemical constituents and pharmacological activities of industry scraps of this plant grown in China were studied systematically to rationally develop and utilize of plant resources and provide more scientific evidence for the development of new drugs.Twenty-one compounds were isolated by methods of column chromatography,thin layer chromatography,recrystallization and high-,medium-pressure preparative chromatography.Their structure were elucidated on the basis of extensive spectroscopic analysis,including HRESI-MS,NMR and IR.Three new compounds were named Liangxiaotane(1),Baccataxinine(2),Baccalignan(3),and eighteen known compounds were determined as Taxinine B(4),Ocinnamoyltaxicin I(5),O-cinnamoyltaxicin I triacetate(6),Taxinine(7),Decinnamoyltaxinine E(8),Taxinine E(9),1β,2α,9α-Trihydroxy-10β-acetoxy-5α-cinnamoyloxy-3,11-cyclotaxa-4(20)-en-13-one(10),5-O-cinnamoyl-9-O-acetylphototaxicin I(11),1-Hydroxyltaxuspine C(12),Taxagifine(13),Taxacin(14),Taxchinin B(15),Taxin B(16),Taxuspine W(17),2α,7β,13α-triacetoxy,5α,9α-dihydroxy-2(3→20)abeotaxa-4(20),11-dien-10-one(18),Taxol(19),Kaempferol(20),Quercetin(21).Compound 1 is a novel skeleton taxanes diterpenoids with a spiral ring.Compound 2 is a new taxane diterpenoids with a 1,3-dioxolane ring at C-1 and C-2.Compound 3 is a benzofuran neolignan.In addition,compounds 8-10,15,16 and 18 were isolated from Taxus baccata L.for the first time.Compounds 1-19 were evaluated for their α-glucosidase inhibitory activity in vitro by the method reported by Nguyen H X,and it was found that there was no significant inhibitory activity.The cytotoxicity of compound 1-3 was investigated against HCT116 and MCF-7 human cancer cell lines by MTT assay,which showed none of them possessed considerable antitumor potency.
