Objective:The effects of Tibetan medicine Lycium ruthenicum M.on oxidative stress induced by exercise fatigue were studied by establishing a model of exercise fatigue in mice.By establishing three free radical generating systems in vitro and three liver microsome lipid peroxidation injury models,the scavenging effect of Tibetan medicine Lycium barbarum on free radicals and the protective effect on lipid peroxidation injury were discussed,so as to provide theoretical basis for further development and utilization of Tibetan medicine Lycium ruthenicum M.?Method:1.150 mice were randomly divided into 3 groups,50 in each group:pre-swim group,immediate after swimming group and 30 min after swimming group?recovery group after swimming?;each group was subdivided into 5 groups,namely:Lycium ruthenicum.M low dose group?3 mg/g?,middle dose group?6 mg/g?,high dose group?12 mg/g?,positive group?red cattle 0.02 mL/g?and blank group?distilled water 0.02 mL/g?10 rats in each group,after continuous administration for 30 days,1 hour after the last administration,swimming for 40 minutes in?25±2?°C water,causing exercise fatigue,then measuring the mice before swimming,immediately after swimming and swimming after 30 min recovery,the serum MDA,LPO,ROS content and SOD,CAT,GSH-Px activity.2.The OH-,O2-,DPPH free radical generation system in vitro was established by kit.The Tibetan medicine Lycium ruthenicum M.reserve solution was prepared with normal saline,the scavenging ability of different concentration of Lycium ruthenicum M.decoction on OH-,O2-,DPPH free radicals were evaluated.3.Liver microsome suspension was prepared from healthy rat liver.Lipid peroxidation injury model was established by three different methods(including CHP,Vc/Fe2+and CCl4/NADP).The effects of different concentrations of Tibetan medicine Lycium ruthenicum M.decoction on the content of malondialdehyde?MDA?in liver microsome lipid peroxidation injury model and its dose-effect relationship were detected.Results:1.There were no significant differences in the contents of MDA,LPO,ROS and SOD,CAT and GSH-Px in the serum of the mice in the swimming group before Lycium ruthenicum M..There was no significant difference?p>0.05?.Compared with the blank group,the medium and high dose Lycium ruthenicum M.aqueous extract can significantly reduce the serum MDA,LPO,ROS content and increase SOD,CAT,and GSH-Px in the immediate after swimming group and 30 min after swimming group.The difference was statistically significant?p<0.05?;and the effects of high dose of Lycium ruthenicum M.were better.2.Different concentrations of Lycium ruthenicum M.have the ability to scavenge O2-,OH-and DPPH free radicals;and as the concentration increases,the scavenging capacity is enhanced.According to the four parameters fitting of ELISACalc software,the half scavenging concentration(IC50)of Lycium ruthenicum M.to O2-was 1.18132 g/mL;the IC50to OH-was 0.94722 g/mL;and the IC50 to DPPH was 1.01458 g/mL.3.In the three lipid peroxidation injury models,Lycium ruthenicum M.with the concentration of 0.6-1.2 g/ml presented a certain extent of inhibitive effects in the lipid peroxidation injury of liver microsomes?p<0.05?,and the effects had a dose-effect relationship.In the VC/Fe2+excitation model,the inhibition rate of Lycium ruthenicum M.?1.2 mg/mL?reached 65%.In the CHP excitation model and CCl4/NADP excitation model,the inhibition rate of Lycium ruthenicum M.?1.2 mg/mL?reached about 50%.Conclusion:1.Lycium ruthenicum M.can significantly alleviate the oxidative stress caused by exercise fatigue,which may be achieved by reducing the serum MDA,LPO,ROS content and increasing the activity of SOD,CAT and GSH-Px in mice with exercise fatigue.2.Lycium ruthenicum M.may alleviate oxidative stress injury caused by exercise fatigue by scavenging free radicals,such as O2-,OH-,DPPH,and thereby increasing the body's antioxidant capacity.3.The decoction of Tibetan medicine Lycium ruthenicum M.can alleviate the lipid peroxidation injury of liver microsomes in a dose-dependent manner.
