Objectives:To explore the establishment of a non human primates?NHP?animal model of spontaneous Alzheimer's disease?AD?and provide an ideal animal model for the study of the pathogenesis of AD.To explore the corresponding relationship between ethology and biomarkers,to verify the feasibility of ethology as an effective method to screen NHP AD model;to compare the correlation between the level of?-amyloid protein??-AP,a??,tau protein in cerebrospinal fluid and the level of a?,tau protein in blood,to verify the feasibility of blood biomarkers as a clinical diagnostic index of AD Take sex.Methods:To select the spontaneous NHP AD model from behavioral and biomarkers,to test the spatial memory ability of experimental monkeys through the delayed response tasks?DRT?,to test the core biomarkers,to collect blood samples and cerebrospinal fluid samples of experimental monkeys by enzyme-linked immunosorbent assay The contents of a?40,a?42 and total tau protein were detected.Results:The results of behavioral test showed that the young group performed better than the old group in the task of short-term delayed response.With the increase of delay time,the difference of memory accuracy between the young group and the old group was more and more obvious,especially in the time difference of 120s delay time?P<0.05?;age and accuracy showed a significant negative correlation,and with the increase of delay time,this kind of The negative correlation was more and more obvious?P<0.05?.The results of biomarker detection showed that a?42,a?40 and a?42/a?40 in serum had no correlation with age,nor with the accuracy of delayed response.The concentration of total tau protein in serum had a greater correlation with the concentration of total cholesterol and high-density lipoprotein cholesterol.Age had a greater correlation with the concentration of total tau protein in CSF and the accuracy of delayed time of 120 seconds There was a significant correlation between a?42/a?40 in CSF and the accuracy of a?42/a?40 in serum and the delay time of 60 seconds.Conclusion:The elderly cynomolgus monkeys showed similar characteristics of possible ad in the delayed response task as the elderly.It can be selected as an ideal model of AD and help to study the pathogenesis of AD.Delayed response task can be used as one of the primary screening methods of AD.the core biomarker of total tau protein in cerebrospinal fluid can be used as the screening index of AD.there is a one-to-one correspondence between the two.Combining the two,it is expected to screen the spontaneous AD model of cynomolgus monkey.Whether the biomarkers in blood can be used as the screening index of AD remains to be further studied.
