| objective:Osteoarthritis(OA)is a kind of chronic progressive joint disease,which involves redness,inflammation,pain,major structural changes of the joint and functional disability.Even in US,people diagnosed with arthritis are projected to increase 49% to 78.4 million.It is said by Global Burden of Disease 2010 study that the global age standardized rifeness of knee Osteoarthritis(KOA)was 3.8% and hip OA was 0.85%,without any obvious change from 1990 to 2010.So,the aim of this study is to evaluate the effects of a novel self-invention fumigation apparatusm-ediated Notch3 and JAG1 in cynomolgus monkey Suffering from Spontaneous Degenerative Knee Osteoarthritis.Methods:Part one:this part employed 16 SPF pour male cynomolgus monkey,which were divided into the aged cynomolgus monkey group A(n=8)and youthy cynomolgus monkey groupB(n=8).Then,we tested with knee circumference,X-ray,ELISA for WBC,IL-1?,TGF-?1,MMP-13 of knee joint fluid and observed articular cartilage(Brittberg score)and Mankin score.Then found out aged cynomolgus monkey Suffering from Spontaneous degenerative knee osteoarthritis.Part two:this study included 15 old pour male cynomolgus monkey(each one aged more than 20 years),which were randomly divided into the new intelligent self-invention fumigation group(A group),the traditional fumigation groupp(Bgroup)and the blank control groupp(C group).Each group has five cynomolgus monkey.Then,the cartilage or soft tissue samples which were taken from interposed monkeys was performed with RT-qPCR and Western Blotting to detect the Notch3?JAG1 and ACAN.Results:Part one:there were seven aged cynomolgus monkeys met with the diagnosis of KOA evaluated by K-L grade and statistical differences in WBC,IL-1?,TGF-?1,MMP-13 between group A and group B(P<0.05).In both Brittberg score and Mankin score group A was statistical differences higher than group B(P<0.05).Part two:With the test of Western-blot,the novel self-invention fumigation and traditional instrument were both worked but the novel self-invention fumigation shows better.the details result as the follows.Notch3,JAG1 were decreased and ANCN increased,Notch3/ beta-actin was:0.36±0.04?0.54±0.02 and 0.83±0.03,(P<0.05),respectively;Jag1/ beta-actin was 0.48±0.01 ? 0.82±0.04 and 1.19±0.06,(P<0.05),respectively;ACAN/ beta-actin was 1.16±0.04?0.84±0.05 and 0.48±0.18,(P<0.05).With the test of RT-qPCR,Notch3,JAG1 were aslo decreased and ANCN increased.Conclusion:Part one:Age,X-ray and ELISA for synovial fluid is likely one of simple but useful methods to choose an aged spontaneous degenerative KOA cynomolgus monkey.Further,the articular cartilaginous Brittberg Scoeand Mankin score test could make a better diagnosis.Part two:the novelselfinvention fumigation could not only effectively inhibit the expression of the knee,Notch3 and JAG1,promoted the expression of ANCN and further but aslo delayed the degeneration of articular cartilage and destruction,so as to effectively delay the arrival of osteoarthritis,which was a small side effect,safe and convenient clinical treatment and trying. |
