Objective:This study was designed to investigate the expression level of TIPE3 in Helicobacter pylori-induced gastroduodenal diseases and to explore the relatioships between TIPE3 and gastroduodenal-related diseases caused by Helicobacter pylori infection.Methods:All experimental cases were selected in Tai'an Central Hospital from January 2016 to January 2017,which were confirmed by gastrointestinal endoscope extraction of gastroduodenal inflammation and gastric cancer paraffin embedded specimens and clinical data.Meanwhile these cases were Helicobacter pylori positive which were confirmed by14C-Urea Breath Test.90 cases were composed of 62 cases of male and 28 cases of female,age between 38-76 years old,mean age 59.2±14.2 years.TIPE3 and CagA expression in tissues were detected by immunohistochemical method in 45 cases of gastric and duodenal inflammation and 45 cases of gastric cancer.The expression levels of TIPE3 mRNA and CagA mRNA were detected by real-time PCR.Western-blots were used to analyse the TIPE3 and CagA protein expression levels in all cases.Results:1.The positive expression rates of TIPE3 in gastric duodenal inflammation and gastric cancer were 48.89%and 82.22%,respectively,which were significantly different from each other.2.The positive expression rates of CagA in gastric duodenal inflammation and gastric cancer were 33.33%and 77.78%,respectively,which were significantly different from each other.3.The expression levels of TIPE3mRNA and CagAmrna were higher in cases with gastic cancer than thoses with gastric duodenal inflammation.4.The expression level of TIPE3 and CagA proteins were higher in cases with gastic cancer than thoses with gastric duodenal inflammation.Conclusion:The expression of TIPE3 and CagA in gastric cancer tissues is higher than that in gastric and duodenal inflammation.Abnormal expression of TIPE3 and CagA in gastric cancer and gastroduodenum inflammation tissues may play an important role in the development of gastric cancer and gastroduodenal diseases caused by helicobacter pylori infection.
