Objective:This study was to investigate the role of FBXW7(F frame/wd-40 domain protein 7)gene in the model of transplanted tumor of esophageal carcinoma cells.Method:Esophageal squamous Eca-109 cells and EC9706 cells were cultured in vitro,and the expression level of FBXW7 gene protein in cells was detected.Lentivirus interference was used to overexpress/knock down the expression of FBXW7 in Eca-109 and EC9706 esophageal squamous cancer cells,respectively.Western blotting were used to verify the transfection efficiency.Finally,the above cells were implanted under the skin of balb/c nude mice to assess the effect of FBXW7 gene expression difference on tumor formation and proliferation of esophageal cancer cells.Result: Tumors were formed in 39 nude mice.In FBXW7 overexpression group,the transplanted tumor grew rapidly.After death,the transplanted tumor was heavier and larger(P<0.01).While the growth of transplanted tumor in FBXW7 knockout group was slower,and the transplanted tumor was lighter and smaller after death(P<0.05).Conclusion: It is suggested that FBXW7 contributes to the formation and proliferation of esophageal squamous cancer cell transplantation tumor.It provides the experimental basis for studying the potential therapeutic targets of esophageal cancer.
