AIM:On the basis of literature research and theoretical discussion,this study designed animal experiments to study the effect of xuanfeiyunpi formula on colon transport function and the expression of 5-HT4,ache,Mn SOD,catalase and GPX protein in colon tissue of mice by establishing STC mouse model,so as to explore the mechanism and dose-response relationship of xuanfeiyunpi formula in the treatment of slow transit constipation The theoretical connotation and the biological mechanism of it can be used as a reference.METHODS:After weighing,50 male ICR mice were randomly divided into two groups:normal control(NC)10 and model control(MC)40.The mice in the model group were made by intragastric administration of 20 mg/kg compound diphenoxylate tablets,while those in the normal group were given equal volume of normal saline once a day for 15 days.After the model was established,the general physiological status and related observation indexes of mice were evaluated to evaluate whether the model was successful or not.Before and after modeling,the weight and gender of mice in each group were analyzed.The models were divided into model group(10 rats),low-dose group(1-yrd),high-dose group(h-yrd)and procapride group(PC).After the treatment of traditional Chinese medicine,the general physiological status and related observation indexes of mice were observed,and the expression of 5-HT4,ache,Mn SOD,catalase,GPX and ?-actin were detected by Western blot.RESULT:1.After the establishment of the model,the total amount of feces in 24 hours in the model group was significantly reduced compared with that in the normal group(**P<0.01),and the carbon ink propulsion rate in the model group was also significantly reduced(**P<0.01).2.Compared with the model group,the total amount of feces and colonic transmission rate increased in 24 hours in xuanfeiyunpi formula group;the expression of 5-HT4,ache,Mn SOD,catalase,GPX and ?-actin protein in xuanfeiyunpi formula group increased compared with the model group,and there was statistical difference.3.The effect of total fecal amount and colon transport rate in 24h in xuanfeiyunpi group was similar to that in pulucapride group;the expression of 5-HT4,ache,Mn SOD,catalase and GPX protein in high dose group was higher than that in western medicine group.CONCIUSION:1.Xuanfeiyunpi recipe can improve the constipation symptoms of STC mice,which is beneficial to the recovery of gastrointestinal motility.2.Xuanfeiyunpi formula can enhance the contraction of colonic smooth muscle in model group by increasing the concentration of 5-HT4 and ache,so as to reduce or treat constipation.In addition,xuanfeiyunpi formula also increased the content of Mn SOD,catalase and GPX,so as to improve the intestinal antioxidant capacity of STC mice,and then improve the defecation function of STC mice.3.The effect of Xuanfeiyunpi formula is similar to that of procapride.
