Objective: The sleep deprivation model rats were prepared by intraperitoneal injection of p-chlorophenylalanine to observe the effects of salidroside on learning and memory functions in sleep deprivation model rats.Methods: Fifty clean adult male Spragus-Dawley rats were selected,and the purchased rats were adaptively fed for 1 week.They were randomly divided into 5 groups: normal control group(C,n=10),and sleep deprivation model group(SDM,n=10),SDM and diazepam injection group(SDM+D,n=10),SDM and low dose salidroside group(SDM+ S1,n=9),SDM and high dose salidroside group(SDM+S2,n=10).Four groups of rats except group C were injected intraperitoneally with parachorophenylalainine(PCPA)for 2 days to prepare a rat model of sleep deprivation.After the model was prepared,all experimental rats were observed and recorded for 24 hours,including weight,diet and water consumption,and circadian rhythm.Diazepam injection(0.92 mg/kg,qd)was injected intraperitoneally into the SDM + D group,SDM + S2 group and SMD + S1 group were injected intraperitoneally with high and low doses of salidroside solution(75 mg/kg,25 mg/kg,qd),group C and SDM groups were injected intraperitoneally with the same volume of normal saline(qd)for 7 consecutive days.On the third day of drug treatment,open field experiments were performed on all experimental rats to detect changes in autonomous activity of each group of rats;and Morris water maze experiment was used to detect the learning and memory function of each group of rats.Results: 1.Comparison of weight at different time pointsThe weight of the rats in each group at the time of enrollment was not statistically significant(F=0.126,P=0.972).The weights of rats in each group were compared before modeling,and the differences in body weight between groups were not statistically significant(F=0.471,P=0.757).After the modeling was completed,the weights of the rats in each group were compared.The results showed that the weight of the rats in group C increased,and the weight of the four groups in the model decreased.The change was significant and the difference was statistically significant(P<0.01).After the treatment,the weight difference between the 4 groups of rats and the C group was statistically significant(P<0.01);compared with the SDM group,the weight difference between the 4 groups of rats was statistically significant(P<0.05).2.Comparison of open field resultsThe horizontal activity of group C rats was higher than that of the other 4 groups.Compared with SDM+D group rats,the difference was statistically significant(P<0.01).Compared with SDM+S2 group rats,the difference was statistically significant(P<0.05),compared with SDM group and SDM+S1 group rats,the difference was not significant(P>0.05).The vertical activity of the SDM group was higher than that of the other 4 groups.Compared with the SDM+D,SDM+S1,and SDM+S2 groups,the difference was statistically significant(P<0.05),but compared with the C group,the difference was not significant(P>0.05).Independent sample t test was performed on the total exercise distance data and central area stay time data of the five groups of rats.The total distance of SDM+D and SDM+S2 groups in the open field box was significantly lower than that of the SDM group(P<0.01,P<0.05).There was no significant difference between the SDM group and the C and SDM+S1 groups(P>0.05).3.Comparison of Morris Water Maze Experiment ResultsTwo-factor analysis of variance(repeated measurement)was used to compare the escape latency of the 7 test days positioning and navigation experiment in each group of rats.The main effects during the test day were very significant(P<0.01)and between the treatment groups(P<0.01);Univariate analysis of variance was used to compare the daily escape latency of each group of rats.The differences between the groups on the second day,the third day,and the fourth day were statistically significant(P<0.01).The time to escape the incubation period was significantly more than the other 4 groups(P<0.05);The independent sample t test was used to compare the experimental groups.The results showed that on the fifth day of the test,the SDM+S2 group took less time to escape the incubation period than the C group and the SDM group(P<0.05).In the space exploration experiment,the target quadrant dwell time in the SDM group was shorter than the remaining 4 groups(P<0.05),and the target quadrant dwell time in the SDM+S2 group was higher than those in the C group(P<0.01)and the SDM+D group(P< 0.05);Compared with the number of platform crossings in SDM rats,the number of platform crossings in SDM+S1 and SDM+S2 groups were significantly different(P<0.05,P<0.01).Among them,the SDM+S2 group crossed the platform the most times,and there was a high statistical difference compared with the C group(P<0.01).Conclusion: The sleep deprivation model rats were replicated by intraperitoneal injection of PCPA,and the changes in body weight,diet and drinking water,and circadian rhythm of each group of rats were observed and recorded for 24 hours to confirm that the experimental model was successfully prepared;The results of open field experiments and Morris water maze experiments showed that salidroside can improve the investigation behavior,spontaneous activity,and calm sleep of rats with sleep deprivation model,and can improve the learning and memory function of rats with sleep deprivation model to a certain extent.
