Purpose: This study investigated the effects of paroxetine at different doses on learning and memory function and induction of long-term synapse enhancement in hippocampal neurons by establishing Obsessive-Compulsiv disorder(OCD)rat models.Methods:Thirty-two adult male SD rats were randomly divided into 4 groups(n = 8):normal control group(C),dark avoidance model group,dark avoidance model given paroxetine low dose group,dark avoidance model given paroxetine high dose Group and M normal control group did not have any treatment,the other three groups were administered by gavage,and the avoidance model was performed by avoiding the darkness model once a day for 15 minutes each time for a total of 7 days.Behavioral testing of rats is an open field experiment,observing and recording the behavior of rats in the open field.After the open field experiment,the normal control group received no treatment.The rats in the dark avoidance group were given saline(1 mg /kg,ig,qd)for 7 days.The other two groups were given paroxetine at a low dose(10mg / kg,ig,qd)and paroxetine in high doses(20 mg / kg,ig,qd)for 7 days.Morris water maze was used to detect learning and memory function.Results:1.Analysis of the results of open field experimentsThe frequency of return home and total exercise distance of the dark avoidancemodel group,the dark avoidance model plus paroxetine high dose group and the dark avoidance model plus paroxetine low dose group before drug intervention were higher than those of the normal control group Group)(P <0.01),the averagereturn time and visiting place were less than the normal control group(group C)(P<0.01 or P <0.05).2.The forced swimming test result analysisThe results showed that the learning effect of rats in each group was significant.The difference(P <0.01),the difference between groups(P <0.05)was statistically significant,The results showed that the learning effect of rats in each group was significant.The difference(P <0.01),the difference between groups(P <0.05)was statistically significant,The main effect during the test day is significant,= 45.12 P<0.05,which indicates that the factor days are statistically significant.The time it takes for the rat to find the platform(escape latency)decreases as the number of testing days increases;the main effect between treatment groups is significant,F =13.243,P <0.05,that is,the group factor has statistical significance,indicating that space learning The escape latency varies significantly between groups.The main effect under the effect of daytime and group two factors was significant,F=1.949,P<0.05,and the pairwise comparison results using SNK-q showed that the escape latency of the normal group was lower than that of the avoidance model group and the paroxetine in the avoidance model.The differences between the dose group and the dark avoidance model plus the high-dose paroxetine group were statistically significant.On the third day of the avoidance incubation period,the avoidance model+ low dose group,avoidance model group was slower than the normal group compared with the normal group(P <0.01),and the avoidance model + high dose group was also slower than the normal group(P<0.05),but faster than the dark avoidance model + low dose group,dark avoidance model group,which has a significant difference compared with the low dose(P<0.05).In the fifth and sixth days of the escape latency,the normal group was faster than the dark avoidance model +low dose group,dark avoidance model group,dark avoidance model + high dose group(P<0.05 or P<0.01),and on the seventh day of the escape latency Compared with the normal group,the dark avoidance model + low dose group,dark avoidance model group,dark avoidance model + high dose group is slower than the normal group(P<0.01),and the dark avoidance model + highdose group is faster than the dark avoidance model + Low dose group,dark avoidance model group(P <0.01).3.Target Quadrant Activity Time PercentageThere were no significant differences in the number of crossings(F= 0.921,P>0.05),and they reached the platform for the first time(F=6.966,P<0.05).After testing SNK-q,a comparison of each pair showed that the normal group was similar to the other three Faster than the first arrival time(P <0.05).Conclusion:1.The obsessive-compulsive disorder model rats induced by the avoidance experiment were tested in the open field and analyzed by the horizontal motion scoring method to verify the success of the model building experiment.2.Paroxetine can improve cognitive function and reduce stereotyped behavior in rats with obsessive-compulsive disorder.3.Paroxetine may have a certain effect on the learning and memory abilities of rats with obsessive-compulsive disorder.
