Objectives:To detect the effects of genistein on proliferation,adhesion,invasion and migration in human thyroid cancer SW579 and TT cell lines,and to explore the possible mechanism of genistein on invasion and metastasis in SW579 and TT cells.To provide a theoretical basis for the application of genistein in the prevention and treatment of thyroid cancer and to find a new way to improve the clinical therapeutic effect of thyroid cancer.Methods:Human thyroid cancer cell line SW579 and TT cells were used as experimental subjects.There were seven groups:positive control group(treatment of normal thyroid follicular epithelial cells Nthy-ori-3-1 with genistein),blank control group and genistein experimental groups(10,20,40,80,160?mol/L).1.SW579 cells,TT cells and Nthy-ori-3-1 cells were treated with different concentrations of genistein for 24h,48h,72h and 96h,respectively;MTT assay was used to detect the effects of genistein on the proliferation activity in the three cells(Nthy-ori-3-1,SW579,TT cells);2.Cell-matrix adhesion assay was used to detect the effect of genistein on the adhesion in SW579 and TT cells;3.Transwell chamber assay was used to detect the effect of genistein on the invasiveness in SW579 and TT cells;4.Transwell chamber method and scratch test were used to detect the effect of genistein on the migration in SW579 and TT cells;5.Real-time fluorescence Quantitative Polymerase Chain reaction(RT-QPCR)was used to detect the effect of genistein on the expression of mmp-2 and mmp-9 mRNA in SW579 cells;6.Statistical methods:SPSS 21.0 software was used to analyze the data.Results:1.Genistein can significantly inhibit the cell proliferation in Nthy-ori-3-1 cells,but significantly promote the proliferation in SW579 and TT cells.In the Nthy-ori-3-1 cells,compared with the control group,the cell survive rate was significantly reduced by all doses of genistein,and there was a dose-response relationship;In the SW579 cells,the number of cells was significantly higher in 80?mol/L and 160?mmol/L genistein groupthan that in the control group(P<0.05);In the TT cell,the cell survive was obviously increased with the increase of genistein concentration,the number of cells was higher in 40 ?mol/L,80?mol/L and 160?mol/L genistein group than that in the control group(P<0.05).2.The cell adhesion was inhibited by Genistein in SW579 and TT cellsCompared with the control group,SW579 and TT cells were pretreated with 160?mol/L genistein for 24 and 48 h,and the cell adhesion was significantly inhibited in SW579 and TT cells(P<0.05).3.The invasion was inhibited by Genistein in SW579 and TT cellsCompared with the control group,the cells passing through Matrigel matrix gel were decreased markedlyin the 40?mol/L group or above.With the increase of genistein concentration,the cells passing through the matrix gel were decreased significantly.Genistein can reduce the invasion in the SW579 cells in this study(P<0.05).In the TT cells,the cells could not pass through the Matrigel into the lower chamber in all groups.4.Migration was inhibited by Genisteins in SW579 and TT cellsIn the Transwell migration experiment,compared with the control group,When the concentration of genistein is 40?mol/L or above,the SW579 cells passing through the Transwell compartment polycarbonate membrane were decreased significantly.With the increase of genistein concentration,the transmembrane cells were decreased.the migration can be reduced by Genistein in SW579 cells in the the study(P<0.05).In TT cells,the cells in all groups were unable to cross the polyester carbonate membrane of Transwell chamber into the lower chamber.In the scratch test,compared with the control group,the blank area of trauma was basically healed in the control group in SW579 cells.At the same time,When the concentration of genistein is 40?mol/L or above,The cells migrating to the blank space was significantly decreased in all genistein groups,and the scratch width was narrowed markedly.In TT cells,the scratch width was no significant change in all groups.6.MMP-2 mRNA expression was inhibited by GenisteinIn the SW579 cells,MMP-2 mRNA was down-regulated markedly by 40,80 and 160?mol/L Genistein(P<0.05),and MMP-9 gene is not expressed.Conclusion(s):1.In this study,the high dose of genistein(80?mol/L and 160?mol/L)significantly promoted the cell proliferation in SW579 and TT cells.However,genistein in medium dose group(40?mol/L)and high dose group(80,160?mol/L)significantly reduced the cell adhesion,invasion and migration in SW579 cells.These results suggested the mechanism of genistein on the cell adhesion,invasion and migration in SW579 and TT cells may be a little different from that in other tumor cells,and further research is needed to be done.2.In this study,genistein may inhibit the adhesion,invasion and migration in SW579 cells by down-regulating the expression of MMP-2.
