BackgroundHigh density lipoprotein?HDL?is well recognized as good lipoprotein in against metabolic syndrome?Mets?,The diversity of HDL lipid composition and structure determines its pleiotropic function. It has many functions such as driven reverse cholesterol transport,anti-inflammatory,antioxidant and others,it plays a very important role in the prevention and treatment of atherosclerosis? As?,and in reducing the risk of coronary heart disease.Past studies have shown that moderate exercise can reduce the incidence of Mets, which improves Mets symptoms and delay complications. The focus of previous treatment on HDL is how to improve the level of HDL-C,But recent research indicate that only increase the level of HDL-C can not effectively reduce the incidence of cardiovascular events. At present, the research direction has shifted to how to improve the function of HDL,This shift may lead to new targets for the prevention and treatment of AS and cardiovascular disease.ObjectiveStudy the effect of square dances exercise on lipid composition and function reconstruction of high density lipoprotein with women metabolic syndrome patients.Methods1.Screening of subjects:The Mets diagnostic criteria is according to the standard put forward by Chinese Diabetes Association in 2004,combind with Chinese commonly used clinical detection project at present, normal exercise capacity is also involved. At last,we screened of the patients with 28 people in metabolic syndrome group and 25 people in control group in line with the conditions.2.Routine blood lipid biochemical detection:fasting venous blood, triglyceride?TG?,total cholesterol?TC?,fasting blood glucose?FPG?,high density lipoprotein cholesterol?HDL-C?and low density lipoprotein cholesterol?LDL-C?.3.Peripheral arteriosclerosis detection:the pulse wave velocity of the two groups were detected by the OMRON arteriosclerosis detector.4.The detection of blood antioxidant:by detection of serum malondialdehyde?MDA?,superoxide dismutase?SOD?,glutathione peroxidase?GSH PX?,catalase?CAT?activity so as to detect blood antioxidant function.5.Vitro cell experiment:through Western blot to measure the expression of cholest-erol ester transfer protein ABCA1 and ABCG1 in HDL and lipid protein apoA1 which was mainly contained in blood and vascular cell adhesion molecule 1?VCAM-1?;Mononuclear cell adhesion assay was observed under the fluorescence microscope, and the effect of HDL on cholesterol efflux was detected by reverse transport of cholesterol.6.Liquid chromatography tandem mass spectrometry detection:determin lipid components and their levels in plasma and HDL?HDL2 and HDL3?.Results1.The clinical features of studies:TC and LDL-C levels of women metabolic syndrome patients in square dances exercise group were lower than that in non exercise group, and the difference was statistically significant, and differences in the indicators of body weight, body mass index, blood pressure, etc.although not statistically significant, but the exercise group of each index were lower than the non exercise group and HDL-C was higher than that in the non exercise group.2.Blood oxidation resistance:square dances exercise group MDA content?4.17±1.04?ml/nmol,significantly lower than the non exercise group content?7.94±2.08?MDA ml/nmol,the difference was statistically significant, but there was no significant difference between the two groups SOD content.Serum CAT activity in the exercise group?56.17±16.32?Ku/l was higher than that of non exercise group?47.38±17.45?Ku/L,and the difference was statistically significant,GSH-PX activity between the two groups had no significant difference. They indicate that square dances exercise may improved antioxidant function of HDL.3.Plasma apoAl level:There was no significant difference in plasma apoAl levels between the two groups.4.HDL3 function:The ability of HDL3 to promote the spillover of cholesterol from cholesterol loaded macrophages in the plasma of the square dances exercise group?0.24±0.38?was significantly higher than the non exercise group?0.08±0.07?,and the difference was statistically significant, and the expression of ABCG1 in exercise group increased and the difference was statistically significant, while the expression of ABCA1had no significant difference,it shows that exercise can improve the ability of HDL to transport cholesterol.the expression of VCAM-1 in the exercise group and the number of single nucleus cell adhesion were significantly less than those of the non exercise group, the difference was statistically significant, which indicated that exercise could improve the anti-inflammatory function of HDL.5.Lipid composition and levels in plasma and HDL:There was no difference in plasma lipid levels between the exercise group and non exercise group of patients with metabolic syndrome;lipids in HDL2 such as SM,C12-C1P?23:0?,C16-CER,C18:1SM,C22-SM,C20-SM,C24-1-SM,DH-SPH,S1P,etc.were significantly different between the two groups;while lipids in HDL3 were amost have significant differences between the two groups, addition to a small portion of such as DH-SPH,SM?18:1/12:0?,C16-CER,etc.It indicated that square dances exercise could improve lipid composition and function of high density lipoprotein in women patients with metabolic syndrome.Conclusions1.Square dance exercise can improve blood lipid levels, lower plasma total cholesterol?TC?and low density lipoprotein cholesterol?LDL-C?levels in women patients with metabolic syndrome.2.Square dance exercise can significantly improve the HDL function, including antioxidant,anti-adhesion and promote the reverse cholesterol transport function in women patients with metabolic syndrome.3.Square dance exercise can significantly improve the lipid composition and function of HDL in women patients with metabolic syndrome.
