BACKGROUNDBreast cancer is currently on a global scale is still a high incidence malignant tumor,although in recent years the treatment of breast tumor had great progress,the fatality rate has been improved significantly than before,but most of the patients with breast cancer diagnosis has been in the middle-late stage,when the late 5 years of survival rate after the conventional radiation and chemotherapy patients still generally lower,therefore,discover new markers for early diagnosis of breast cancer and prognosis assessment is particularly important Bioinformatics is a with the rapid development of computer science,and the birth and life science combined to form a new discipline,this study using bioinformatics tools and methods with a huge number of network database data extraction in breast cancer high-throughput gene sequencing results are retrieved and inductive analysis,mining potential key genes associated with breast cancer,the research in the pathogenesis of breast cancer and targeted drugs research and development to provide a new train of thought.AIMTo to screen Hub genes related to breast cancer and analyze the relationship between biological function and prognosis by bioinformatic analysis methods and tools.METHODSTwo microarray datasets were downloaded from Gene Expression Omnibus(GEO)database.The differentially expressed genes between breast cancer samples and normal tissue samples were screened using GEO2R tools.The functional and pathway annotations of the differentially expressed genes were performed by Gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses,Meanwhile,the protein-protein interaction network(PPI)was constructed using STRING and Cytoscape to pick up the hub genes,and the relations of the hub genes with prognosis were analyzed using the Kaplan-Meier plotter database.Finally,the protein expression and distribution of hub genes in breast cancer tissues and normal tissues were verified again according to the immunohistochemical results in the HPA database.RESULTSA total of 127 differentially expressed genes were screened,of which 36 were up-regulated and 91 were down-regulated.Ten HUB genes such as AURKA CDK1 PCNA were obtained by Cytoscape software analysis.The survival analysis showed that except the CDK1 SMC,the abnormal expressions of other hub genes were significantly associated with the overall survival of the breast cancer patient.CONCLUSIONSThe abnormal expressions of the screened hub genes may be involved in the development and progression of breast cancer,and could be closely related to the prognosis of breast cancer patients.These genes may serve as potential predictors and therapeutic targets for further research of breast cancer.
