Objective: In this study,we used DART to establish a rapid and simple method for qualitative and quantitative analysis of Basic Auramine O and Orange ? in bean products.We introduce a novel ESI method that uses Sunflower seed husk for loading and ionization of samples.Methods: In this study,DART-Q-Exactive Orbitrap mass spectrometry was used to establish a rapid and simple method for qualitative and quantitative analysis of the fruits of Basic Auramine O and Orange ? in bean products.The Full MS/Targeted MS2 mode and internal standard method were applied to qualitative analysis of these components.Additionally,we set up a simple ESI device which using Sunflower seed husk for loading and ionization of samples.And it is positioned nearly the inlet of mass spectrometer and the sharper point to that.It's well load sample and there is electrospray produced at the tip of Sunflower seed husk when high potential is applied.The method can be used for detecting targeted ion without sample prepared.And comparisons of wooden tip spray with sunflower seed husk(WTSSSH)with ESI and wooden tip spray with toothpick in terms of analytical performance was provided in the manuscript.Results: In the study of qualitative and quantitative analysis of the Basic Auramine O and Orange ?.The mass spectrum profile was obtained and the fragmentation pathways of the two pigments were investigated.Under positive ion mode,the quantitative result of Auramine O and Orange ? was obtained by choosing basic schisandrin as an internal standard.The method showed good linear coefficient-in the concentration range of 1~20?g/m L of Auramine O and Orange ?.The correlation coefficients were 0.9994 and 0.9972,respectively.The limit of detection was 0.2 ? g/m L.In this manuscript,we report a simple and economical type of electrospray ionization(ESI)technique,which makes use of disposable wooden tips of sunflower seed husk for loading and ionization of samples.Samples could be loaded by simply dipping the tip into sample solutions or normal pipetting onto the natural groove of Sunflower seed husk.Instrument parameter settings were optimized to obtain the sensitive and high ion intensity determination of samples;The wooden tip of the Sunflower seed husk was pointed to the mass spectrometer inlet for the Q-Orbitrap mass spectrometers,and there are optimum angles between 60? and 90? in wooden tip.The tip was positioned 2~3mm away from the inlet of mass spectrometer.The applied voltage to wooden tip of Sunflower seed husk was adjusted in the range of 3.5~4k V until optimal ion signals were obtained and all desolvation gases were turned off.Ideal mass spectrometric information can be immediately.Comparisons of wooden tip spray with sunflower seed husk(WTSSSH)with ESI and wooden tip spray with toothpick were investigated with lignin,ginsenoside and proteins.The quality of Mass spectra by using WTSSSH was better than other two techniques.Some special sample,e.g.powder,can't be directly analyzed by ESI and wooden tip spray with toothpick,but it is convenient for WTSSSH.In this study,we established a continuous injection device which was designed according to the single needle injection method of ESI.The ginseng powder was online extracted with the device and extracts was ionized in wooden tip of sunflower seed husk for mass spectrometric analysis,and a continuous and stable total ion chromatogram and abundant information of mass spectra were obtained.Conclusion: The method was applied successfully for the qualitative and quantitative analysis of Basic Orange ? and Auramine O in bean products.The experimental results showed that this new ionization technique is applicable for analysis of various samples,e.g.lignin,ginsenoside, aminoacid,proteins,etc.The method is rapid and accurate,and can provide high throughput screening samples and inspecting illegal addition of pigments in foods.And it could provide valuable information and indicating the potential applications in traditional Chinese medicine analysis,clinical and forensic analysis.
