Objective: To establish a morphological identification method and a molecular identification method by studying six plants of Angelica L.in the northeast three provinces,and to provide scientific basis for accurately identifying these plants.In this study,the contents of total phenols,total flavonoids,total sugars,and antioxidant activities in different solvent extracts of Angelica sinensis root and Angelica dahurica root were systematically studied.And the volatile oil components and biological activities of Angelica Sinensis root,Angelica gigas root and Angelica dahurica root were also studied.The above studies lay the foundation for their development and utilization.Methods: 1.We collected six plants of Angelica L.and described their morphological characteristics by comparing them with the description of "Chinese Flora".Sequence screening by DNA barcode identification.2.The extracts of the plant were obtained by reflux extraction method,and the content of total phenol,total flavonoid and total carbohydrate in each extract was determined by spectrophotometry.DPPH,ABTS,FRAP,CUPRAC,carotene bleach method were used to evaluate the antioxidant activity of plant extracts,and MTT method was used to evaluate the cytotoxicity of the extract.3.The GC-MS technology was used to analyze the plant volatile oils.The hypoglycemic activity of the extract was evaluated using ?-glucosidase and ?-amylase.The anti-cholinesterase activity of the extract was evaluated using acetylcholinesterase and butyrylcholinesterase.Results: 1.Through morphological comparison,the morphological characteristics of six plants of Angelica L.were confirmed.Through the study of DNA barcode,the ITS2 sequence can achieve the best identification effect.2.Seven solvent extracts were obtained after systematic extraction of Angelica Sinensis root.The experimental results showed that the methanol layer has the highest total phenol and total flavonoid content,and the water extract has the highest total carbohydrate content.methanol and ethanol layer showed high DPPH and ABTS scavenging ability,they also showed high reducing ability in FRAP and CUPRAC experiments.Chloroform and water extracts showed better ability to scavenge hydroxyl radicals.Ethyl acetate extract could effectively prevent ?-carotene bleaching and cell oxidative damage.Water and methanol extracts showed lower cytotoxicity.Study on the Extract of Angelica dahurica root,The experimental results showed that the methanol extract has the highest total phenol content,and the acetone extract has the highest total flavonoid content.In the study of antioxidant activity,the methanol extract showed high DPPH scavenging ability and iron ion reducing ability,acetone extract showed high ABTS scavenging ability,and ethanol extract showed strong copper ion reducing ability.Hexane extract is effective in preventing ?-carotene bleaching.3.The GC-MS technology was used to analyze the components of volatile oil from the roots of Angelica Sinensis,Angelica gigas and Angelica dahurica.38,71 and 72 components were identified respectively.The results of the activity experiments showed that the volatile oil of Angelica Sinensis and Angelica gigas had certain antioxidant activity and Hypoglycemic activityConclusion: 1.The effective identification characteristics of each plant of Angelica L.in the northeast three provinces were summarized through the morphological identification.A systematic evaluation of 6 plants using 4 fragments of DNA barcode sequences was performed at the molecular level.and the IST2 fragment played a key role in identifying Northeast Angelica species.2.A systematic study of Angelica Sinensis root and Angelica dahurica root showed that the extracts from each layer have certain activity.3.38,71 and 72 components were identified from the volatile oils of Angelica Sinensis,Angelica gigas and Angelica dahurica respectively.Studies on the biological activity of volatile oil from Angelica Sinensis root and Angelica gigas root showed that both volatile oils have certain antioxidant activity,good ?-glucosidase inhibitory activity.These results provide detailed data for their research and development and comprehensive utilization.
