For the systematic comparison of active substances and developing new medicine source of Angelica Miq. plants, we studied on the chemical constituents of A.acutiloba(Sieb.et.Zuce.) Kitag. for the first time and systematically studied their active ingredients, including total coumarins, voltatile oils and decursin on the basis of chemotaxonomy.We studied the chemical constituents of the extract of 85% ethanol from the A.acutiloba(Sieb.et.Zuce.)Kitag. and 5 compounds were isolated. Their structures were confirmed as bis(5-formylfurfuryl)ether(1), 5-hydroxymethyl-2-furaldehyde(2), stigmaasterol(3), heptadecanoic acid (4) by spectrums, and mixture of ester(5) by GC-MS.1~4 compounds were isolated for the first time from A.acutiloba(Sieb.et.Zuce.) Kitag., together with compound 1 was isolated for the first time from Angelica Miq. Plants.The contents of the total coumarins of 8 species of Angelica Miq. plants for the first time by UV, and the results show that the content of A.decursiva f. albiflora. is the highest, A.gigas Nakai. is followed. The best condition of UV:coumarin was the comparison,95% ethyl alcohol was the resolver, wavelength was 274 nm and the method was accurate, rapid, simple and easy to use, could be used as the one methods of quality control of Angelica Miq. Plants.The method of GC-MS-DS was used to identify the chemical components of the essential oils from 8 kinds of Angelica Miq. plants, and the common compounds were mainly terpenoids. The composition of volatile oils of A.peucedanum decursivun (Miq.) Maxim. and A.gigas Nakai.'flower were very similar and the compound type is more than root far. Meanwhile, we have compared simultaneous distillation extraction and water-steam distillation method and the results show that, SDE retained more low boiling point components than water-steam distillation.RP-HPLC method was established for quantitative analysis of decursin for the first time and the best condition of HPLC is that wavelength was 280 nm, temp was 30℃, flow rate was 1 ml/min, mobile phase was PH=5.0, H3PO4-NaH2PO4(dilution 10 times):CH3CN(1:1). The method was simple, fast and could gain good accuracy, suitable for quantitative analysis of decursin in Angelica Miq. Plants.The results provided the important theoretical evidence for chemical composition exposition, appications and brought into national standards of A.acutiloba (Sieb.et.Zuce.) Kitag. It also provided important theoretical foundation for identification of active ingredients, chemical classification and search for new drug source from Angelica Miq.Plants. Thus, it is worthy of developing national herbs and modernization of Traditional Chinese Medicine.
