BackgroundIn recent years, the quality of medical care and the development of science and technology have been continuously improved, which has significantly increased the detection rate of prostate cancer, and it has penetrated the entire elderly population in society.Prostate cancer is a high-risk malignant tumor in the male population, which is very harmful.More and more, there are more and more patients diagnosed with prostate cancer, especially the detection rate of patients with early prostate cancer has increased significantly. But overall, the number of these patients with advanced prostate cancer is still far more than the early screening of prostate cancer patients. The overall treatment plan for prostate cancer includes endocrine therapy, laparoscopic prostate cancer resection and robotic laparoscopic prostate cancer resection, etc. These therapies can effectively treat prostate cancer. However, patients with advanced prostate cancer, especially those with bone metastases. After 2-3 years of endocrine therapy, most of them will turn into castration-resistant prostate cancer.(CRC)There is no better treatment plan, and the prognosis of CRPC patients is generally worse. Therefore,it is very beneficial to study the progression mechanism of prostate cancerWe discovered earlier that a gene B3GALT2 is likely to play an important role in the progression of prostate cancer. However, the expression of B3GALT2 in prostate cancer and its role in prostate cancer have not been reported.B3GALT2 is a member of the beta-1,3-galactosyltransferase(beta3GalT)gene family. This family is able to encode with different types of donor substrates(UDP-galactose and UDP-N-acetylglucosamine)and different kinds of acceptor sugars(N-acetylglucosamine,galactose,N-acetylgalactosamine)Type ? membrane-bound glycoprotein with multiple enzyme functions. This gene plays a role in the glycosylation of N-linked glycoproteins, and shows that the expression of B3GALT2 in prostate cancer and its mechanism of action in tumors have not been reported. In this study,we used bioinformatics methods to analyze the expression and function of B3GALT2 in prostate cancer, and combined with molecular biology methods to verify,in order to further discover the mechanism of action of B3GALT2 in prostate cancer, transform it into a new Molecular targets provide a new basis for the diagnosis and treatment of prostate cancerObjectiveUsing bioinformatics analysis methods to explore the difference between B3GALT2 expression in normal prostate and prostate cancer tissues, and to study the impact of B3GALT2 on the survival of prostate cancer patients. To study the biological role of B3GALT2 in prostate cancer, and provide new targets and treatment strategies for the diagnosis and treatment, repeated detection, and prognosis of prostate cancer through biological informationMethod:1.Download the prostate cancer project in the TCGA dataset and analyze the project data to study the expression of B3GALT2 in prostate cancer and the impact of B3GALT2 expression on survival2.Using qRT-PCR,WB detected the expression of B3GALT2 mRNA and protein in prostate hyperplasia cell line BPH1 and prostate cancer cell line LNcaP and PC33.After knocking down and overexpressing B3GALT2 in the prostate cancer cell line LNcaP,CCK8 was used to detect the proliferation ability of prostate cancer cells, the prostate cancer scratch test method was used to detect the cell migration ability, and the flow cytometry method was used to detect the cells The cyclical change proves that B3GALT2 plays an important role in cancer suppression and genome suppression in the treatment of prostate cancerResult1.Bioinformatics data show that the expression level of B3GALT2 in normal prostate cancer tissue is significantly higher than that in prostate cancer tissue2.Prostate cancer patients with low B3GALT2 expression have a poor prognosis for progression-free survival3.The expression level of B3GALT2 RNA decreased in BPH1,LNCaP and PC 3 cell lines in turn At the protein level, the expression of B3GALT2 protein was consistent with the expression of RNA The expression level in BPH1,LNCaP and PC3 decreased in turn.4.The methods of qRT-PCR and Western Blot proved that after transfecting siB3GALT2,the expression levels of B3GALT2 RNA and protein in LNCaP cells were significantly reduced.Similarly, after transfection of the B3GALT2 overexpression plasmid, the expression levels of B3GALT2 RNA and protein in LNCaP cells increased significantly5.CCK8 experiment shows that B3GALT2 has the effect of inhibiting the growth of prostate cancer.6.The scratch experiment further confirmed that B3GALT2 can effectively inhibit the migration inhibition ability of malignant prostate cancer.7.The experimental results of flow cytometry used to detect cell cycle show that B3GALT2 cells can effectively guide the inhibition of cell cycle arrest.8.Through GO analysis and analysis of the correlation of cell expression,we can find that B3GALT2 may work through genes such as GNL3,TRAF4,HNRNPL,PTBP1,CDK16,LETM1,RUVBL1 and other genes.Conclusion1.This subject found that B3GALT2 is low expressed in LNcap cells through PCR,flow cytometry,Western blot,cck8 and other research detection methods, and its low expression promotes LNCaP cell proliferation.2.At the same time,B3GALT2 was knocked out and overexpressed in LNcap cells, and it was found that high expression of B3GALT2 inhibited the migration ability of prostate cancer cells.3.B3GALT2 provides a possible exploration method for the study of prostate cancer.
