| Objective: To compare the effect and relationship between IL-6 and TGF-?1 in promoting the transformation of synovial cells into chondrocytes,and to explore the role of both in the pathogenesis of synovial chondromatosis.Method: In the experimental group,the synovial tissues from the bursa suprapatellaris was isolated and cultured in a sterile environment.When the synovial cells were passaged to the third generation,the purity of the synovial cells was detected by flow cytometry.The experimental teams was divided into group A,group B,group C,group D,group E and group F.each group has added pre-configured liquid.there were 9 specimens in each group.On the 2nd and 4th day of culture,3 samples were taken from each groups for type ? collagen immunofluorescence detection,SOX9 immunofluorescence detection and proteoglycan ELISA detection.The immunofluorescence intensity was measured by Image J software,and the intra-group and inter-group results of each group were compared.Results: The intensity of immunofluorescence of type ? collagen shows that,the group with 100ng/mlIL-6+sIL-6R+10ng/ml was higher than that in the other 5 groups(P < 0.05),100ng/mlsIL-6R+10ng/ml TGF-?1 and 10ng/ml TGF-?1 were higher than those in the other three groups(P< 0.05),100ng/mlIL-6+sIL-6R and 100ng/ml IL-6R + 10 umol / ml TGF-?1 blockers were higher than group A,the blank groups(P < 0 05).What's more,there was no significant difference between 100ng/ml sIL-6R+10ng/ml TGF-?1 and 10ng/ml TGF-?1 in the intensity of immunofluorescence,as soon as the intensity of immunofluorescence between 100ng/mlIL-6+sIL-6R and 100 ng / ml IL-6R + 10 umol / ml TGF-?1 blockers(P > 0.05).The analysis of the intensity of immunofluorescence of SOX9 and the expression of proteoglycan have shown the same results.Conclusions : Both TGF-?1 and IL-6 can promote the transformation of knee joint synovial cells into chondrocytes,but the promoting effect of TGF-?1 is stronger and there may be no obvious relationship between them. |
