| Objective: Recently,researchers have found that lipopolysaccharide(LPS)promotes the development and metastasis of a variety of cancers,including breast cancer,but the specific mechanism is not fully understood.The purpose of our study was to investigate whether LPS can promote the proliferation,migration,and invasion of MDA-MB-231 cells and its possible mechanisms.Methods: MDA-MB-231 cell lines were routinely cultured,and the experiment was divided into 5μg/ml group,10μg/ml group,20μg/ml group,40μg/ml group and the control group without adding LPS according to the concentration of LPS solution.The cell proliferation experiment,cell scratch experiment and cell invasion experiment were conducted in groups,and the expressions of VEGF-R2 and VEGFD in cell lines of different concentrations were detected by ELISA kit.Results: In the experiments of cell proliferation,scratch and invasion,the MDA-MB-231 cell lines stimulated by LPS showed an obvious trend of proliferation,migration and invasion compared with the control group,and the trend was positively correlated with the concentration of LPS.The difference was statistically significant(P < 0.05).In addition,the expressions of VEGF-R2 and VEGF-D in the five experimental groups gradually increased with the increase of LPS concentration,and the differences between each two groups were statistically significant(P < 0.05).Conclusion: LPS promoted the proliferation,migration and invasion of MDAMB-231 cells,.The expression levels of VEGF-R2 and VEGF-D in MDA-MB-231 cells were positively correlated with the concentration of LPS,which suggesting that the role of LPS in promoting the development of MDA-MB-231 cells might be related to the formation of blood vessels and lymphatics... |
