| Objective:Traditional Chinese medicine granule is a new kind of preparation,which is easy to carry,easy to take,easy to store and keep the curative effect of traditional Chinese medicine.However,there has been a lot of controversy since the introduction of formula granules,which focuses on whether they can really replace traditional decoction,and whether they can match traditional decoction in composition,content and efficacy.Pinellia xiexin decoction is a famous classic prescription in traditional Chinese medicine,with a history of more than one thousand years.There is a lot of clinical experience to prove that this prescription is simple and effective,and it is a commonly used prescription in traditional Chinese medicine.However,there is no literature on the comparison of traditional decoction and formula granules in pinellia xiexin decoction.It remains to be seen whether the convenient formula granules can be used in place of traditional decoction in the clinical use of pinellia xiexin decoction.The composition and content of pinellia xiexin decoction were determined in this experiment.It is hoped that it can play a guiding role in the clinical use of pinellia xiexin decoctionMethods:1.Referring to the content determination method in Chinese Pharmacopoeia,the content of index components in Scutellaria baicalensis,Coptis chinensis,ginseng,prepared liquorice,dried ginger was determined,and the content of index components of formula granules from different manufacturers was different According to the fingerprint of each single drug,the peak situation was compared,including the difference of peak number,peak type and common peak content,so as to further compare the ingredient difference of formula particles of scutellaria baicalensis geoli,Coptiana,ginseng,prepared liquorice and dried ginger.2.On the basis of the content determination method of "Chinese pharmacopoeia",a suitable chromatographic method for the determination of baicalin,berberine hydrochloride,palmatine,coptidine,rhizoma corydalis,glycyrrhizic acid,6-gingerol was established by many experiments.The content difference between traditional decoction and granule in pinellia xiexin decoction was compared according to the chromatogram.Results:1.Composition comparison between traditional decoction pieces and formula granules.Comparison results of Scutellaria baicalensis:Among all manufacturers,the content of scutellaria baicalensis formula granules was the highest in Manufacturer A(3.72%).Comparison results of Rhizoma Coptidis:according to the calculated alkaloid contents of rhizoma coptidis,the highest contents were berberine 3.00%,bamatine 0.81%,rhizopine 0.62%and rhizopine 0.67%respectively.Comparison results of processed glycyrrhizae granule:Among the formula granule of processed glycyrrhizae produced by manufacturer A,the content of glycyrrhizin was the highest,0.62%.Manufacturer B has the most glycyrrhizin content,which is 0.97%.Comparison results of dried ginger:In the comparison of formula granules from various manufacturers,the content of 6-gingerol in formula granules from Manufacturer A was the most,which was 0.08%.Ginseng comparison results:Ginsenoside Rg1 content comparison:Among the formula granules from various manufacturers,the content of Ginsenoside Rg1 in the formula granules from Manufacturer A was the most,0.12%.Comparison of ginsenoside Re content showed that ginsenoside Re content of formula granule B from various manufacturers was the highest,0.21%.Comparison of ginsenoside Rbl content:Formula granules from Manufacturer B had the most content(0.45%).2.Results of composition comparison between traditional decoction and formula Granule of Pinellia TernataComparison results of index components:Baicalin:the content of traditional decoction is the most.The content of baicalin in the traditional decoction was 0.416g/agent,and the content of Formula granule was 0.176g/agent.Berberine hydrochloride:the content of traditional decoction is the most.The content of traditional decoction containing berberine hydrochloride is 0.064g/agent,and the content of formula granule of various manufacturers is higher in A,0.052g/agent.Paracetamol:The most common form is the traditional form.The content of bamatine in traditional decoction is 0.019g/agent.Compared with formula granule of various manufacturers,the content of A is higher,0.014g/agentRhizoma Coptitis:the content of traditional decoction is the most.The traditional decoction contains 0.016g/agent of Coptis line.Compared with the formula granule of various manufacturers,the content of A is higher,0.010g/agentRhizocine:the content of traditional decoction is the most.The traditional decoction contains 0.011g/agent,and the content of D in formula granule of various manufacturers is higher,0.006g/agentGlycyrrhizin:The granule content of manufacturer B is the largest.The content of glycyrrhizin in the formula granule of Manufacturer B is 0.0436g/agent,while the content of glycyrrhizin in the traditional decoction is 0.0304g/agent6-Gingerol:Traditional decoction contains the most.The traditional decoction contains 0.0114g of 6-gingerol per agent,while the content of formula granule of various manufacturers is higher,0.0041 g/agentComparison of peak emergenceUnder various chromatographic conditions,the traditional decoction had more peaks than formula granules,and most of the common peaks had higher content Under the chromatographic condition of determination of glycyrrhizin,the common peak content of no.12 and no.14 was lower than that of formula granules.Under the chromatographic condition of determination of 6-gingerol,the content of no.2 common peak decoction was lower than that of formula granulesConclusion:By parallel comparison,it was found that there was a general difference in the content of ingredients measured by different manufacturers.In the comparison of pinellia Xiexin decoction,it was found that the composition and content of the granule were generally lower than the traditional decoction.The results provide a basis for unifying the quality standard of formula granule. |
