Objective:Traditional Chinese medicine formula granules are products of the new era.They have the advantages of quick dispensing,no decocting and convenient carrying,etc.The use rate of patients is increasing year by year.Therefore,whether its composition,component content,and pharmacological effects can replace traditional decoctions,has gradually become a hot topic of discussion and research by traditional Chinese medicine workers.Zexie decoction is derived from Golden Cabinet(Jingui Yaolue)and selected into the Classic Catalogue of Ancient Classics(First Batch).It is widely used in clinical practices and has definite curative effect.At the same time,Zexie decoction is a basic prescription,and a large number of clinical prescriptions are modified from Zexie decoction.Nonetheless,there is no related research on the comparison between traditional decoction and formula granule of Zexie decoction.In this experiment,quantitative analysis was performed by measuring the extract rate and the content of index components,efficacy analysis was performed by establishing a cell model,to compare the difference between traditional decoction and formula granule of Zexie decoction.With a view to provide a reference for the reasonable clinical application of Zexie decoction formula granule.Methods:1.To simulate the traditional decoction process,the formula granules were dissolved and decocted.Then the difference in the extract rate and the content of index components between different decoction time formula granules and traditional decoctions were compared.2.Three batches of melt samples,decoction samples and traditional decoctions in the extract rate and the differences in the content of index components were compared.3.HepG2 cells were randomly divided into control group,Zexie decoction formula granule dissolution sample group,Zexie decoction formula granule decoction sample group,and Zexie traditional decoction group.MTT method was used to compare the effects of different concentrations on cell viability.4.HepG2 cells were induced by oleic acid to establish fat accumulation model.The absorbances of Zexie decoction formula granule dissolution sample group,Zexie decoction formula granule decoction sample group,and Zexie traditional decoction group were compared.To evaluate the inhibitory effect of the three on cell fat accumulation.5.The difference in the extract rate and the content of index components between the decoction samples and the melted samples of the Zexie decoction formula granules from five manufacturers were compared.6.The difference of the extract rate and the content of index components of the different manufacturers of Zexie decoction formula granules were compared.7.The differences in the content of index components of Zexie decoction,Alismatis Rhizoma and Atractylodes Macrocephala were compared.Results:1.The contents of 23-acetylalismol B,23-acetylalismol C,Atractylenolide ?,and Atractylodes ? in samples of Zexie decoction formula granules decocted for 5 minutes were higher than those of Zexie decoction formula granules for 2 minutes,10 minutes sample.The similarity between the characteristic spectrum of Zexie decoction formula granules decocted for 5 minutes and that of the Zexie traditional decoction was high.And the similarity between the characteristic spectrum of Zexie decoction formula granules decocted for 5 minutes and that of the Zexie decoction formula granules melted samples was high.2.There were differences in the content of the index components of the decoction sample,melted sample,and traditional decoctions.But it had similar effects on inhibiting lipid accumulation inHepG2 cells.3.The extraction rate and the content of index components of the five manufacturers of decoction sample were generally higher than those of the melted sample.4.Among the five manufacturers,A manufacturer's extraction rate,23-acetylalismol B,Atractylenolide ?,and Atractylenolide ?are higher.5.The contents of 23-acetylalismol B and 23-acetylalismol C in the Alismatis Rhizoma sample were generally higher than those of Zexie Decoction sample.Conclusion:In this study,it was found that there were differences between the dissolved samples,decocted samples and traditional decoction of Zexie decoction,the extraction rate and the content of index components.But it had similar effects on inhibiting lipid accumulation inHepG2 cells.There were differences in the extraction rate of the granules of the Zexie Decoction from different manufacturers,the contents of 23-acetylalismol B,23-acetylalismol C,atractylodes lactone ?,and atractylodes lactone ?.This study provides reference for the rational use of Zexie Decoction granules.Providing research methods for the consistency evaluation between formula granules and traditional decoctions of Zexie Decoction.To provide ideas for quality control and standard formulation of Alismatis Rhizoma and Atractylodes Macrocephala formula granules.
