| The chemical components of medicinal plants and the secondary metabolites of microorganisms from animals and plants are one of the sources of natural drugs.In this paper,we isolated and identified the chemical components of the lipid-soluble part of the famous Chinese herb Balanophora harlandii in Meizhou area of Guangdong Province.Combined with the activity and chemical screening methods,four symbiotic microorganisms were selected to isolate and identify the chemical components of its secondary metabolites.The main contents are as follows:(1)Four candidate strains were identified by morphological characteristics and ITS/16S rDNA gene sequence analysis.Strain Mzz9 belongs to Penicillium,It is 99.9%similar to the sequence of Penicillium coprophhilum.Strain BM6 belongs to Aspergilus sp.,It is 99.9%similar to the sequence of Aspergillus clavatus and Aspergillus clavatonanicus.Strain TY1 belongs to Streptomyces sp.,Strain XM3 belongs to Penicillium,It is 99.9%similar to the sequence of Penicillium citrinum.(2)28 compounds were isolated,identified from Balanophora harlandii and 4 symbiotic microorganisms selected by using multiple chromatographic and spectroscopic.Five compounds were isolated from the dichloromethane extracts of the Chinese herb,Balanophora harlandii:lupeol(1),β-amyrin(2),clearesterol(3),lupeol acetate(4),glycerin linoleate(5);Penicillium cophilum Mzz9,an endophytic fungus isolated from the stem of Balanophora harlandii,Six compounds were identified:dechlorinated griseofulvin(6),griseofulvin(7),dehydrogenated griseofulvin(8),oxaline(9),4-megastigmen-3,9-dione(10),p-hydroxyacetophenone(11).Six compounds were identified from Strain Aspergillus sp.BM6 isolated from zebra feces:clavato(12),3-hydroxybenzoic alcohol(13),o-methylhydroquinone(14),Z-N-(4-hydroxy styryl)formamide(15),Theobroxide(16)and patulin(17).Four compounds were identified from strain Penicillium sp.XM3 isolated from panda feces.Their structures were 2,4,5-trimethyl benzene-1,3-diol(18),citrinin(19),dicitrinin A(20),penicillinol A(21).Sevene compounds were identified from Strain Streptomyces sp.TYl isolated from alpaca feces.Their structures were N-[2-(4-hy droxyphenyl)ethyl]acetamide(21),N-acetylserotonin(23),4-hydroxyphenylethanol(24),methyl 4-hydroxybenzoate(25),3,4-dihydroxy-4-methyl-2-pentanone(26),N-[3-(1H-indole-3-yl)propyl]acetamide(27),indazole(28)(3)Patulin,the compound 17 from Aspergillus sp.BM6 was resistant to S.aureus,E.coli,F.oxyaporum and C.albicans.The MIC values are 16,8,16 and>64μg/ml,respectively.Some compounds were tested for MDCK cytotoxicity.Compounds 14,18 and 20 showed a certain toxicity to MDCK.Their CC50 were<25,<25,25-50 μg/mL respectively,while other compounds had no significant toxicity to cells within 50 μg/mL. |
