Mercury,lead,and chromium are three of the most important inorganic poisons in forensic toxicology analysis.Mercury,lead,and chromium are widely distributed in nature,and they can accumulate in living organisms.Because of their different compounds,the toxicity varies widely.Organic toxicity is tens to hundreds of times that of inorganic toxicity.In view of the different toxicity of different compounds of mercury,lead,and chromium,it is necessary to establish a highly sensitive morphological analysis method to determine the content of mercury,lead,and chromium in human blood and urine in the field of forensic toxicology analysis.Blood and urine are commonly used as samples for analysis of human mercury,lead,and chromium.Based on the content of mercury,lead,and chromium in biological samples,human exposure to mercury,lead,and chromium can be evaluated.because of their simple interfaces,lower detection limits,wider linear range,relatively few interferences,simple pre-processing procedures,and help keep the original shape of the sample under test,high-performance liquid chromatography-inductively coupled plasma-mass spectrometry(HPLC-ICP-MS)analysis method is widely used in elemental morphology analysis.As showed below,the contents of this study can be divided into 3 parts.Chapter 1:Analysis of Mercury Compounds in Blood and Urine by HPLC-ICP-MSTo establish an analysis method for the detection of 3 mercury compounds[methyl mercury,ethyl mercury,phenyl mercury]in blood and urine by HPLC-ICP-MS.Benzene was used to extract mercury compounds,sonication and sodium thiosulfate were performed for sample pretreatment.With the mobile phase consisted of ammonium acetate and methanol,gradient elution was performed for obtaining the mercury compounds in samples,which were analysed by ICP-MS with C,8 column.The limits of detection in blood and urine were 0.96-1.67ng/mL,the lower limits of quantitation in blood and urine were 5.00ng/mL,the RSD was 0.6%-6.3%,the recovery rate was 88.3%-102.1%,and the matrix effect was 98.3-?113.4%.Chapter 2:Analysis of Lead Compounds in Blood and Urine by HPLC-ICP-MSTo establish an analysis method for the detection of 2 lead compounds[trimethyllead,triethyllead]in blood and urine by HPLC-ICP-MS.Benzene was used to extract lead compounds,sonication and sodium thiosulfate were performed for sample pretreatment.With the mobile phase consisted of ammonium acetate-ammonium and methanol,gradient elution was performed for obtaining the mercury compounds in samples,which were analysed by ICP-MS with C18 column.Results The limits of detection in blood and urine were 0.96-1.50ng/mL,the lower limits of quantitation in blood and urine were 3.00-5.00ng/mL,the RSD was 0.6%-6.3%,the recovery rate was 88.3%-102.1%,and the matrix effect was 98.3%-113.4%.Chapter 3:Analysis of Chromium Compounds in Urine by HPLC-ICP-MSTo establish an analysis method for the detection of 2 chromium compounds[chromium ?,chromium ?]in blood and urine by HPLC-ICP-MS.Samples were prepared with bufer solution containing EDTA,with the mobile phase consisted of 65%HNO3 and deionized water(DIW),the chromium compounds were separated by Bio-WAX column which were analysed by ICP-MS.The limits of detection in blood and urine were 0.25-0.30ng/mL,the lower limits of quantitation in blood and urine were 1.00ng/mL,the RSD was 4.7%-9.8%,the recovery rate was 80.8%-91.0%,and the matrix effect was 84.1%-88.4%.
