Objective:The present study was designed to investigate the relationship of the expression of vascular endothelial growth factor(VEGF)and cyclooxygenase 2(COX-2)with the clinicopathological characteristics and angiogenesis in cancer tissue in patients with invasive ductal breast cancer.Methods:Paraffin blocks from 100 cases of cancer tissues and 30 cases of paracancerous tissues resected or punctured in Yunnan Cancer Hospital from 2016 to 2019,and 40 cases of breast benign lesions who underwent minimally invasive surgery were selected.The expression of VEGF and COX-2 in breast invasive ductal carcinoma,adjacent tissues and breast benign lesions was detected by immunohistochemical method(immunohistochemistry,IHC).The correlation between them and the clinicopathological characteristics of breast cancer patients were analyzed.In addition,the microvascular endothelial cells of breast invasive ductal carcinoma were labeled with CD34 antibody,the microvessel density(microvascular density,MVD)was calculated,and the MVD of VEGF and COX-2 under different expression conditions were compared.Results:1.The expression rates of VEGF and COX-2 in breast invasive ductal carcinoma were 73%and 72%,respectively,which were significantly higher than those in adjacent tissues(36.67%and 30%)and benign breast lesions(45%and 37.5%),respectively(P<0.005).2.In invasive ductal carcinoma of the breast,the high expression of VEGF and COX-2 was found in the cancer tissues with primary focus>2cm(P<0.05),and the higher the histological grade and the later the TNM stage was,the higher the expression of VEGF and COX-2 was,and the difference was statistically significant(P<0.05).There was no significant difference in the expression of VEGF and COX-2 among different ages and molecular types(including the comparison between triple-negative and non-triple-negative invasive ductal carcinoma of the breast)(P>0.05).3.There was a significant correlation and positive correlation between the expression of VEGF and COX-2 in breast invasive ductal carcinoma(r=0.875,P<0.005).4.In breast invasive ductal carcinoma,the MVD of patients with positive expression of VEGF and COX-2 was significantly higher than that of negative expression of VEGF and COX-2(P<0.001),and the MVD of patients with positive expression of VEGF and COX-2 was significantly higher than that of negative expression of VEGF and/or COX-2(P<0.001).Conclusion:1.The expression of VEGF and COX-2 in breast invasive ductal carcinoma was significantly higher than that in adjacent tissues and benign breast lesions.2.The expression of VEGF and COX-2 in breast invasive ductal carcinoma increased with the increase of primary focus size,histological grade and TNM stage,and the difference was statistically significant.There was no significant correlation between the expression of VEGF and COX-2 and the age and molecular classification of patients(including the comparison between triple-negative and non-triple-negative invasive ductal carcinoma of the breast).It is suggested that the high expression of VEGF and COX-2 in breast invasive ductal carcinoma may be closely related to the malignant degree of breast cancer,and they may promote the development and metastasis of breast cancer.3.The MVD of VEGF and COX-2 positive expression was significantly higher than that of VEGF and COX-2 negative expression,and the difference was statistically significant;and the MVD of VEGF and COX-2 positive expression was significantly higher than that of VEGF negative and/or COX-2 negative expression,the difference was statistically significant;in addition,correlation analysis showed that there was a significant correlation and a significant positive correlation between the expression of VEGF and COX-2 in breast invasive ductal carcinoma.It is suggested that both of them may play a synergistic role in the neovascularization of invasive ductal carcinoma of the breast.
