Objective: This study was performed to investigate the effect of Nocardia rubra cell wall skeleton(N-CWS)on wound healing of full-thickness skin defects.Methods: Twelve healthy adult male SD rats weighing 200 ? 250 g were excised with scalpel at the symmetrical parts of the left and right sides of the rat's back with a total of 24 full-thickness skin defects.Using self-control method,the wound on the right side of the midline was used as the control group,covered with vaseline gauze soaked in normal saline,and fixed with gauze.The wound on the left side of the midline was used as the N-CWS treatment group,and was externally applied with 0.02% N-CWS2 ml,and fixed with vaseline gauze and gauze.The wound dressing was changed every 2days,and the wound size was recorded.The wound healing time and the wound healing rate on the 10 th day were observed.On the 7th day after injury,4 rats were killed and wound specimens were taken.Immunohistochemistry was used to observe the expression changes of CD31,CD68 and F4/80,and Western blot was used to observe the expression changes of transforming growth factor(TGF)-?1.Results: The wound healing rate on the 10 th day after injury in rats was(89.555.68)% in the N-CWS treatment group and(86.363.23)% in the control group,which was statistically significant(P=0.026).The time for complete wound healing was(11.131.63)days in the N-CWS treatment group and(12.881.02)days in the control group,which were statistically significant(P=0)On the 7th day after injury,immunohistochemical analysis showed that the expression of CD68 in the wound surface was(9705.031602.79)(IOD/area)in theN-CWS treatment group and(4777.671292.44)(IOD/area)in the control group,which was statistically significant(P=0.004)The expression of F4/80 in the wound surface was(3413.20781.99)(IOD/area)in the N-CWS treatment group and(2886.78550.77(IOD/area))in the control group,which was statistically significant(P=0.025)On the 7th day after injury,Western blot results showed that the wound TGF-?1,N-CWS treatment group was(0.610.05),while the control group was(0.410.04),with statistical significance(P=0.001)The expression of CD31,a specific marker of endothelial cells,was(7489.151063.75)(IOD/area)in the N-CWS treatment group and(4975.851027.36)(IOD/area)in the control group,which were statistically significant(P=0.001)Conclusions:1.N-CWS can improve the healing rate of skin defect wound for 10 days and shorten the time of complete wound healing.2.N-CWS increases the expression of CD68 and F4/80 in the wound surface,activates the macrophages in the wound surface,increases the expression of TGF-?1,and enhances angiogenesis,thus promoting the healing of full-thickness skin defect wound surface.
