Turmeric?Curcuma longa L.?is a perennial herbaceous plant in the Zingiberaceae family.Curcumin is a natural active substance of polyphenols,mainly found in the rhizome of turmeric.Curcumin has various pharmacological effects such as anti-inflammatory,anti-viral,anti-oxidation,anti-tumor,and treatment of neurological diseases.It has good clinical application prospects,but it has properties such as poor water solubility,low bioavailability,and short half-life,which limit its development and application.This study aims to solve the shortcomings of low solubility and low bioavailability of curcumin.Through the compatibility of turmeric with licorice,astragalus and angelica,the curcumin content and biological activity of turmeric after compatibility are analyzed.The solubilization of curcumin and the inhibitory effect of curcumin combined with cisplatin on cancer cells determine the best ratio of curcumin to cisplatin,and then prepare curcumin composite nanoliposomes and evaluate the biological activity.The application of nanoliposomes provides a certain theoretical basis,and the specific research results are as follows:1.Research on the biological activity of turmeric.When turmeric is combined with licorice,angelica and astragalus,the ratio of curcumin of the three compatibility reaches the maximum when the ratio is 1?1.2,and the content of curcumin is 8.79mg·g-1,7.08 mg·g-1 and 7.44 mg·g-1,respectively.When turmeric compatible with licorice 1?0.4,compatible with angelica 1:1.6,compatible with astragalus 1?1.6,the removal rate of DPPH·and ABTS·+free radicals reached the maximum,of which turmeric compatible with licorice 1?0.4,the antioxidant activity is highest,and the free radical scavenging rates for DPPH·and ABTS·+are 89.58%and 83.27%,respectively.The ratio of turmeric to licorice is 1:2,the ratio of angelica is 1?1,and the ratio of astragalus is 1?2,which has a good effect on the inhibition rate of Bacillus subtilis and Staphylococcus aureus.Among them,the best anti-bacterial ability of turmeric Compatible with turmeric and astragalus,the relative bacteriostatic rate was 42.89%and 28.34%.Turmeric and licorice compatibility ratio is 1?0.4,compatibility angelica ratio is 1?1,compatibility astragalus ratio is 1?2,the proliferation inhibition rate of breast cancer cell MDA-MB-231 and lung cancer cell H460 is good,Among them,the anti-tumor ability of turmeric and licorice was the best,and the cell suppression rate was 41.67%and 51.08%.2.Curcumin solubilization and its combined cisplatin antitumor activity.The results show that:when it is at 10 mmol·L-1,glycyrrhizic acid can maximize the solubility of curcumin,which is 607 times higher than the control;under 25?water bath and 50%ethanol can promote the dissolution of curcumin in glycyrrhizic acid solution;the critical glue of glycyrrhizic acid The beam concentration is 0.65mmol·L-1.Both curcumin and cisplatin have an inhibitory effect on the proliferation of liver cancer cell HepgG2,and the inhibition rate continues to increase with increasing concentration;the inhibitory effect of cisplatin and curcumin on the proliferation of liver cancer cell HepgG2 increases,the ratio of the two is 1?8,The best inhibitory effect on the proliferation of liver cancer cells HepgG2.3.Study on the biological activity of curcumin composite nano-liposomes.The thin film dispersion method,ethanol injection method and reverse evaporation method are used to prepare curcumin composite nanoliposomes.The particle size distribution prepared by the thin film dispersion method is more uniform,presents a regular spherical shape,and the dispersion degree is better;The particle size of the liposome is 359 nm,the potential is-46.81 mV;the liposomes prepared are regular in shape and have good encapsulation efficiency.The biological activity of curcumin-composite nanoliposomes showed better inhibitory effect than ascorbic acid,when 100?g·mL-1,liposomes had the strongest antioxidant effect,and the scavenging rates of DPPH·and ABTS·+free radicals were 53.89%and 97.89%;when 1000?g·mL-1,curcumin composite nanoliposome has the best comprehensive bacteriostatic effect,and the inhibition rates against Escherichia coli and Bacillus subtilis are 18.00%and 21.88%,against wisteria pink and banana The inhibition rates of the disease were 10.67%and 33.56%;when 12.5?mol·L-1,the proliferation inhibition rates of curcumin composite nanoliposomes on liver cancer cells HepG2and lung cancer cells H460 were 86.67%and 78.98%,respectively.
