Danshen,the dried root or rhizome of Salvia miltiorrhiza Bge.,is a traditional and folk medicine in China.And it is used to treat various pains caused by blood stasis,lumps in the chest and abdomen,pyogenic infection and carbuncle of the skin,palpitation and insomnia.Many compounds of Danshen have been summarized so far.These chemical constituents are divided into two groups based upon their structures: lipophilic diterpenoids and water-soluble phenolic acids.In practice,two main active components of Salvia miltiorrhiza Bge.are obliged to extracted by different methods for their opposite properties.which results in the waste of resources.The pressure solvent extraction technology was applied for the extraction of Salvia miltiorrhiza Bge.in this paper,in order to simultaneously extract two active components of Salvia miltiorrhiza Bge.,furthmore,extraction kinetics was studied for improve the comprehensive utilization of resources of Salvia miltiorrhiza Bge.The research contents and results are as follows:1.The effects of different extraction methods on the yield of Salvia miltiorrhiza Bge.extract were studied.The extract components were qualitatively analyzed using high performance liquid chromatography(HPLC),and the activity of the extracts by different extraction methods was evaluated by pharmacodynamics studies.Compared with the traditional water boiling method and ethanol reflux method,the extraction rate of pressurized solvent was the highest,which was 43.52 %.In vivo pharmacodynamic evaluation showed that the extracts from different methods all have certain increase the hypoxia tolerance and anti-fatigue effects in mice;in vitro antioxidant experiments,the scavenging effect of DPPH and ·OH and the reduction capacity of iron ions showed that the pressure extraction method had higher antioxidant activity than the traditional extraction methods at the same concentration.Although pressurized solvent extraction is extracted under the high temperature and high pressure,but won't destroy drug ingredients.2.The active components of Salvia miltiorrhiza Bge.were extracted by pressure solvent extraction.The effects of extraction solvent concentration,extraction temperature,extraction time,solid-liquid ratio and pressure on the extraction rate of Salvia miltiorrhiza Bge.were investigated by single factor experiment.On this basis,the extraction rate of Salvia miltiorrhiza Bge.extracts,total ketone extraction rate of Salvia miltiorrhiza Bge.and total phenolic acid extraction rate of Salvia miltiorrhiza Bge.were used as the indexes for optimization of extraction conditions,by adopting the method of star design-response surface,the optimal extraction process is obtained as follows: the concentration of ethanol was 70.2 %,the extraction temperature was 134.22 ?,and the extraction time was 84.17 min.The lipid soluble and water-soluble components of Salvia miltiorrhiza Bge.could be extracted simultaneously under the conditions of the extraction process.The lipid soluble and water-soluble components of Salvia miltiorrhiza Bge.could be extracted simultaneously under the conditions of the extraction process.3.The mass transferred mechanism of extraction of Salvia miltiorrhiza Bge.with pressurized solvent was studied.The effects of temperature on the extraction rate and mass transfer process of total ketones and total phenolic acids were analyzed.The established second-order kinetic equation:Total ketones:(?)Total phenolic acids:(?)It can be used to describe the mass transfer process of extraction with pressurized solvent and provide a theoretical basis for industrial production of extraction of active components of Salvia miltiorrhiza Bge.4.The process cost of traditional water decoction,ethanol reflux,pressurized water extraction and pressurized alcohol extraction were discussed,which enriched the application prospect of pressurized solvent extraction in traditional Chinese medicine extraction.In conclusion,pressurized solvent extraction technology with high extraction efficiency,environmental support benefits and power conservation,and can simultaneously extract the two active ingredients in Salvia miltiorrhiza Bge.,which opens a new direction for the modernization of traditional Chinese medicine.
