Objective: Salvia miltiorrhiza water extraction liquid of exploration and discovery with cadmium intervention of kidney damage in rats, to explore its possible mechanism of action, and to expand the salvia miltiorrhiza water extraction liquid cadmium to kidney injury intervention research results provide a scientific basis for widely used in clinic.Methods: Choose healthy Wistar rats, male and female each 20. Adaptive feed after 1 week, were randomly divided into two groups: blank control group, model group, respectively. 32 only eight blank control group, model group, conventional breeding, intraperitoneal injection of saline blank control group 5 ml/kg.d-1, model group to 2.8 mg/kg.d-1intraperitoneal injection of cadmium chloride solution(that is,the preparation of cadmium chloride solution 5 ml/kg.d-1). Building four consecutive weeks, observe the general situation of rats, weight change, detection rat renal function indexes, urine β2-MG, MDA, SOD and kidney tissue pathological changes.To build after the success of the model, model group were randomly divided into the drug group(salvia miltiorrhiza water extraction liquid of high dose of 2.7 g/kg.d-1,salvia miltiorrhiza, water extraction liquid of low dose of 1.35 g/kg.d-1), negative control group, positive control group. Dosage group lavage with salvia miltiorrhiza water extraction liquid, and positive control group using sodium selenite solution 0.05mg/kg.d-1 lavage, negative control group, blank control group given volume normal saline lavage with 2.7 ml/kg.d-1. Each intervention treatment for 10 weeks, to the terminal to collect serum, urine, and to return the kidney tissue, detection in the serum creatinine, blood urea nitrogen, SOD, MDA, serum total antioxidant capacity(T-AOC), urine β2-MG, renal cortex and urine, blood cadmium content, kidney pathological tissue damage, changes of serum trace elements, and the detection of renal cortical cells apoptosis rate.Results: The experimental results showed that salvia miltiorrhiza water extraction liquid group compared with blank control group no difference rat kidney weight coefficient, salvia miltiorrhiza water extraction liquid high dose groupcompared with negative control group with significant difference; Salvia miltiorrhiza water extraction liquid of low and high dose group of 24 h urine significantly less than the negative control group, the serum SOD increased significantly, serum MDA is larger, higher total antioxidant capacity, blood urea nitrogen, serum creatinine content decreased significantly, the urine β2-MG content decreased obviously; Kidney tissue pathology slice HE dyeing result shows the negative control group in renal proximal convoluted tubules epithelial cells turbidity, swelling, degeneration, even have cell necrosis, disintegration, small changes, such as tube cavity stenosis; Salvia miltiorrhiza water extraction liquid of low and high dose group of serum levels of cadmium and cadmium content is significantly lower than the negative control group:renal cortex can be adjusted to dye cadmium rats serum zinc, iron, calcium and other trace elements of imbalance; Renal cortical cell apoptosis rate showed that salvia miltiorrhiza water extraction liquid of low and high dosage group and positive control group, apoptosis rate is lower than the negative control group.Conclusions:( 1) Salvia miltiorrhiza water solution can accelerate the metabolism of cadmium and the cadmium toxicity to kidney injury have better intervention effect. Its mechanism may be related to regulating serum zinc, iron,calcium and other trace elements of imbalances.(2) the salvia miltiorrhiza water extraction liquid cadmium cause kidney damage, can effective intervention helps delay the chronic renal failure and improve renal function, protect the kidney. Its mechanism may be related to salvia miltiorrhiza water extract has stronger removal of oxygen free radicals and antioxidant injury, inhibiting cell apoptosis, and other functions are closely related.
