The Mechanism And Clinical Effect Of Kuntai Capsule In The Treatment Of Premature Ovarian Failure

Objective:To explore the mechanism and clinical effect of Kuntai capsule in the treatment of premature ovarian failure,so as to provide theoretical basis for clinical treatment of premature ovarian failure.Methods:From January 2018 to June 2019,90 patients with premature ovarian failure were randomly divided into three groups: combined group(n=30),group A(n=30)and group B(n=30).According to the order of treatment,90 patients with premature ovarian failure were randomly divided into three groups: combined group(n=30),group A(n=30)and group B(n=30).Drug administration: Kuntai capsule 4 tablets,3 times a day,half an hour after meal.Fenmoitong 1 tablet / d,every 28 days for 1 cycle,the first 14 days,daily oral administration of 1 white tablet(14 days after containing 1 mg);of estrodiol,daily oral administration of 1gray tablet(containing 1 mg of estrodiol and 10mg),3 cycles as a course of treatment.After one course of treatment,the results of the three groups were recorded and statistically analyzed.The changes of TCM syndrome score,inhibin B,follicular stimulating hormone(FSH),estrogen(E2)and mean ovarian volume(MOV)were observed before and after treatment.The expression of antioxidant enzyme(SOD2),apoptosis-related protein Bax,Bcl2 and anti-Mule tube hormone(AMH)in blood of three groups was detected by West blot.Result:(1)Comparison of TCM syndrome score and curative effect before and after treatment:there was significant difference in the total score of TCM syndrome between the three groups before and after treatment(P<0.05),and the total score of TCM syndrome in the combined group and the total score of TCM syndrome in treatment group B after treatment were compared,the results showed that the advantage of the combined group was more obvious,and the difference was statistically significant(P<0.01).After treatment,the total effective rate of TCM syndrome in combined group was 86.7%,and that in group A was70.0%.The total effective rate of treatment group B was 66.7%.There were significant differences in the curative effect of the three groups in improving TCM syndromes,amongwhich the combined group had the best curative effect,the difference was statistically significant(P<0.05).(2)Comparison of the improvement of TCM syndromes before and after treatment: the combined group was significantly superior to the other two groups in improving menstrual cycle,menstrual period,insomnia,weakness of waist and knee,dizziness and tinnitus,hand and foot heart fever,hot and cheekly red,etc.The results of the combined group were significantly better than those of the other two groups in improving menstrual cycle,menstrual period,insomnia and dreams,waist and knee weakness,dizziness and tinnitus,hand and foot heart fever,hot and cheekly red and so on(P<0.05).(3)Comparison of biochemical indexes before and after treatment: after treatment,the basic biochemical indexes of the three groups were compared and compared with those before treatment.The levels of inhibin B,FSH,E2 and MOV were improved before and after treatment,among which the three groups were superior to those before treatment,and the results were statistically significant(P<0.05).Compared with the other two groups,the levels of inhibin B,FSH,E2 and(MOV in the combined group were significantly higher than those in the other two groups(P<0.01).(4)Comparison of the expression of SOD2,Bax,Bcl2,AMH protein: the expression of SOD2,Bax,Bcl2 and AMH in the blood of the three groups were detected.The results showed that the expression levels of SOD2,Bax,Bcl2 and AMH in the three groups after treatment were significantly higher than those before treatment(P<0.05).The expression levels of SOD2,Bax,Bcl2 and AMH in the combined group were significantly higher than those in the other two groups(P<0.05).Conclusion:(1)Kuntai capsule combined with fimotong in the treatment of premature ovarian failure can effectively improve the symptoms of TCM syndromes,improve the therapeutic effect of patients,and significantly improve the symptoms of premature ovarian failure.(2)Kuntai capsule combined with fimotong can effectively improve the menstrual cycle,menstrual volume,symptom effect and endocrine level of patients with premature ovarian failure.(3)Kuntai capsule combined with fimotong can significantly improve the levels of inhibin B,follicular estrogen,estrogen and mean ovarian volume in patients with POF.(4)Kuntai capsule combined with fimotong can increase the level of Bax,Bcl2,increase and the content of SOD2 in blood and enhance the expression of AMH in patients with premature ovarian failure.
