Objective:Experimental study clearly Jiaji electroacupuncture Effects of spinal cord injury IL-18 and IL-1? and motor function,the inhibition clarify Jiaji EA IL-18 and IL-1?,decrease inflammation in spinal cord injury,by the use of clinical Jiaji electroacupuncture treatment of spinal cord injury provide experimental and theoretical basis.Methods:The experiment was divided into two parts.Experiment 1: To study the effects of Jiaji electroacupuncture on limb motor function in SCI rats.120 female SD rats were randomly divided into sham operation group,model group,Jiaji electroacupuncture group,NLRP3 interference group,NLRP3 interference control group,24 rats in each group,each component was divided into 4 time points(1d,3d,7d,21d),6 rats at each time point.Sham operation group: only expose the spinal cord without any other treatment,postoperative routine feeding,only tied together with other groups of rats;model group: routinely raised after successful modeling,tied together with other groups of rats,and the rest Any treatment;Jiaji electro-acupuncture group: Acupuncture treatment will be given to rats 3 hours after successful modeling,until the end of each course of treatment.NLRP3 interference group: lentivirus injection after successful modeling.NLRP3 interferes with the control group: rats with successful ASCI models were injected with NLRP3 negative control lentivirus.The BBB score was used to evaluate the motor function of rats in each group.Experiment 2: Observe the effect of Jiaji Electroacupuncture on the expression of pro-inflammatory factors IL-18 and IL-1? in the spinal cord tissue of SCI rats.The experiment grouping is the same as experiment 1.They were given appropriate disposal or treatment.Expression of rat spinal cord tissue by IHC IL-18 and IL-1?.To clarify the effect of Jiaji Electroacupuncture on the expression of proinflammatory factors in spinal cord tissue ofSCI rats.Result:(1)BBB scoreBBB scores showed that at 1d,3d,7d,and 21 d postoperatively,compared with the sham operation group,the BBB scores of the model group,NLRP3 interference group,NLRP3 interference control group,and Jiaji electroacupuncture group all decreased significantly,and the difference Statistically significant(P<0.05);At 3d,7d,and 21 d postoperatively,the BBB scores of the model group,NLRP3 interference group,NLRP3 interference control group,and Jiaji electroacupuncture group all showed an upward trend,with the highest score at 21d;At 3d,7d and 21 d,compared with the model group,the BBB score of the Jiaji electroacupuncture group increased significantly,and the difference was statistically significant(P<0.05);compared with the NLRP3 interference control group,the NLRP3 interference group BBB score Significantly increased,the difference was statistically significant(P<0.05).(2)IHC detects the expression of IL-18IHC test results showed that compared with the sham operation group,IL-18 expression in the spinal cord tissue of each group of rats at each time point was significantly increased,the difference was statistically significant(P<0.05),indicating that the model was successfully prepared;compared with the NLRP3 interference group,At 3d,7d,and 21 d,NLRP3 interfered with the increase of IL-18 expression in the spinal cord tissue of the control group.The difference was statistically significant(P<0.05),indicating that the construction of the interfering lentiviral vector was successful;after treatment with Jiaji electroacupuncture,3d,7d,21 d Jiaji electroacupuncture group spinal cord tissue IL-18 expression were lower than the model group,the difference was statistically significant(P<0.05);plasmid virus loaded with si RNA interfered with the expression of NLRP3,3d,7d,21 d The expression of IL-18 in the spinal cord tissue of rats in the NLRP3 interference group at each time point was lower than that in the model group,and thedifference was statistically significant(P<0.05).(3)IHC detects the expression of IL-1?IHC test results showed that the expression of IL-1? in the spinal cord tissue of each group was higher than that of the sham operation group at each time point,the difference was statistically significant(P<0.05);NLRP3interfered with the control group rats at 3d,7d,and 21 d The expression level of IL-1? in the spinal cord tissue was higher than that in the NLRP3 interference group,and the difference was statistically significant(P<0.05);At 3d,7d,and 21 d,the expression of IL-1? in the spinal cord tissue of the Jiaji electroacupuncture group was lower than that of the model group.,the difference was statistically significant(P<0.05);After the plasmid virus loaded with si RNA interfered with the expression of NLRP3,the expression level of IL-1? in the spinal cord tissue of rats in the NLRP3 interference group at 3d,7d and 21 d was lower than that of the model group,the difference was statistically significant Significance(P<0.05).Conclusion:1.Jiaji electro-acupuncture can improve lower limb motor function of SCI rats and promote the recovery of nerve function.2.Jiaji electroacupuncture can inhibit the expression of inflammatory factors IL-18 and IL-1? in the tissue,improve the microenvironment,and inhibit the inflammatory damage of SCI rat tissues.
