Sanghuang is the fruit body of Phellinus igniarius,which is a fungus belonging to Polyporaceae,Polyporales.The main chemical components of P.igniarius are polysaccharides,steroids,terpenes,and so on.The pharmacological activities of this fungus include antitumor activity,immunomodulatory activity,anti-cirrhosis activity,and so on.We took Sanghuang as our research object.Through modern chromatography separation technologies and structure identification techniques such as NMR spectroscopy and mass spectrometry,the chemical comstituents were systematically separated and identified.Then we evaluated these compounds' anti-neuroinflammatory activity,laying a solid foundation for the discovery of anti-neuroinflammatory lead compounds.The P.igniarius was pulverized,and then added to MeOH for refluxing abstraction.After the abstraction,ethyl acetate was added for extraction.Then preparative HPLC and different kinds of column chromatography,such as AB-8,MCI CHP20 P,silica gel,Toyopearl HW-40 F,ODS,Sephadex LH-20,were used to isolate and purify compounds.The structures of the isolated compounds were analyzed by NMR spectroscopy and mass spectrometry.BV-2 cells were used to evaluate the anti-neuroinflammatory activity of the compounds.Nineteen compounds were isolated and identified from Sanghuang,including five new compounds: Phellinols A-D(1-4),N-((5-phenyloxazol-2-yl)methyl)acetamide(5),with fourteen known compounds: protocate-chualdehyde(6),(E)-4-(3,4-dihydroxyphenyl)-3-buten-2-one(7),inoscavin A(8),davallialactone(9),interfungins A(10),phelligridins F(11),phelligridimer A(12),methyl protocatechuate(13),methyl caffeate(14),phellibaumins A(15),(22E,24R)-ergosterol-7,22-diene-3?-one(16),(22E,24R)-ergosterol-7,22-diene-3?-ol(17),2,2'-oxobis(1,4-ditertbutylbenzene)(18),cerebroside B(19).Additionally,compounds 7,8,12,14,18 and 19 displayed an inhibitory effect against the lipopolysaccharideinduced nitric oxide production in BV-2 microglial cells.
