Study On The Chemical Constituents And Biological Activity Of Chuanshan Orange

Firstly,the paper reviewed the research progress of bisindole alkaloids from natural products based on the previous reported literatures.Secondly,we carred out the work around isolation and structure identification of chemical compositions from Melodinus tenuicaudatus,especially for alkaloids.Thirdly,the immunosuppressive,anti-inflammatory and cytotoxicity activity of all alkaloid compounds were evaluated.Finally,we conclued all the theories and practices in research process.The genus Melodinus belonging to the Family of Apocynaceae consists of approximately with 53 species.There are about 11 species that found in China,used as folk medicines for treatment some diseases.The phytochemical investigation on the aerial parts of M.hemsleyanus led to the isolation and identification of 4 new compounds along with 33 known compounds,including seventeen tabersonine type alkaloids(1-17),three vincadine type alkaloids(18-20),six quinolone type alkaloids(21-26),five vincamine type alkaloids(27-31)and six bisindole alkaloids(32-37).Meanwhile,in this thesis by lymphocyte transformation test in mice,these compounds which were isolated and identified from Melodinus hemsleyanus were researched of immunosuppressive activity.The results showed that the compounds 6,9,12,37 exhibited exceptionally potent immunosuppressive activities at the concentration of 40 ?M.Compounds 11,20,22,25 displayed significant immunosuppressive activities at the concentration of 20 ?M while the compounds 16 and 18 at the concentration of 10 ?M.By studying the effects of triterpenes and some pregnane glocosides on the release of IL-6 and TNF-? from mouse macrophages,found that compounds 6 and 32 have anti-inflammatory activity.The cytotoxic activity of the isolated alkaloids on human tumor cells was tested.The results showed that the compound 4 have significantly inhibited the activity of the two human tumor cells.
