Coastal Glehnia Root is the dried root of Glehnia littoral& Fr. Schmidt ex Miq. (Faro. Umbelliferae). It’s used in traditional Chinese medicine for the treatment of respiratory and gastrointestinal. It has traditional actions including replenishing yin of the lung and stomach, removing heat from the lung, and promoting fluid secretion. Previous investigations resulted in the isolation of coumarins, polyacetylenes, lignans, phenolic acids, and nucleotides etc. On the other hand, it has been reported that this crude drug exhibited many pharmacological activities, including antitumor-promotion, autimutation, antitumor, antibacterial and antifungal, analgesicand sedative, inhibition of tyrosinase, and immunosuppressive activities.In this study, chemical constituents of the roots of G. littoraIis and its anti-gastric ulcer activity were investigated.The roots of G. littoralis were extracted with ethanol, and the extract was suspended in water, and then partitioned with pertroleum ether and EtOAc, successively. The H2O-soluble fraction was chromatographed on macroreficular resin column, repeated Sephadex LH-20 and reverse phase ODS column chromatography to give eleven compounds. Their structures were elucidated by physieo-chemical properties and spectral analysis. Among them, one new coumarin glycoside was named bergaptol 5-O-β-D-genfiobioside. Ten known compounds were identified as (7R, 8S)-dehydrodiconiferylalcohol-4, 9-di-O-β-D-glucopyranoside (2), cirtrusin A (3), (-)-secoisolarici resinol 4-O-β-D-glucopyranoside (4), glehlinoside A (5), ferulic acid (6), baihuaqianhuside (7), xanthotoxol 8-O-β-D-glucopyranoside (8), 5’-methylthioadenosine (9), adenosine (10), L-tryptophan (11). Compound 2 and 9 were isolated from this genus for the first time.Furthermore, the anti-gastric ulcer activity of H2O-soluble fraction was evaluated using the HC1/EtOH induced gastric ulcer in mice, which displayed 38.5% inhibition with 400 mg/kg (i.g.), P<0.01. Then we examined the hydroxyl radical scavenging activity of H2O-soluble fraction, which showed potent activity (IC50 = 0.48 mg/mL), and the H2O-soluble fraction was reported to be potent in inhibiting lipid peroxidation. Thus we suppose that its mechanisms of anti-gastric ulcer may be accelerating scavenging of free radicals in gatric mucosa and relieving lipid peroxidation.
