Objectives to construct the model of miRNA-449 a mimics and inhibitors to explore their effects on the migration and proliferation of human colon cancer cells LoVo and SW480,so as to provide partial data for the study of miR-449 a at the cellular level of colon cancer,and to provide some theoretical basis for early diagnosis,effective treatment and prognosis evaluation of colon cancer.Methods Human colon cancer cell lines HT-29,HCT-15,LoVo,SW480 and human normal colon epithelial cell NCM-460 were cultured,and the expression level of miRNA-449 a in each group was detected by real-time quantitative PCR.According to the results,two groups of cells,LoVo and SW480,were selected and cultured and divided into 4 groups: transfection of miR-449 a mimics,miR-449 a mimics,NCmiR-449 a inhibitor and miR-449 a inhibitor NC respectively to construct miRNA-449 a mimics and inhibitor models and their corresponding NC mimics.After the successful construction of the model,the proliferation of cells was detected by CCK-8 kit and the ability of cell migration was detected by cell scratch test to explore the effect of miRNA-a449 on the migration and proliferation of LoVo and SW480 in human colon cancer cells.Western blot assay was used to detect the expression of proliferation-related antibody protein ?-catenin and migration-related antibody protein E-Cadherin.Results compared with normal human colon epithelial cells,the expression of miR-449 a was low in human colon cancer cells HT-29,HCT-15,SW480 and LoVo,and the lowest expression was found in SW480 and LoVo cells.After the construction of miRNA-449 a mimic model,it was found that the proliferation and migration ability of colon cancer cells LoVo and SW480 decreased,while the proliferation and migration ability of colon cancer cells increased in the miRNA-449 a inhibitor model.The results of Western blot assay showed that the expression of proliferation-related protein ?-catenin and migration-related protein E-Cadherin was inhibited in the mimic model,while the expression of proliferation-related protein ?-catenin and migration-related protein E-Cadherin was enhanced in the inhibitor model.Conclusions transfection of miRNA-449 a mimics or inhibitors can affect the proliferation and migration of colon cancer cells to some extent,and the effect is more significant on LoVo cells.Figure 8;Table 20;Reference 170...
