Objectives By establishing the(MS)rat model of metabolic syndrome,the effects of Shuangqing Pinghua Formula on abdominal obesity,blood lipids,blood glucose and insulin resistance(IR)in MS rats were observed,and the mechanism of regulating blood lipids was discussed.Methods 60 SD rats were fed adaptively for one week,according to Excel function,the rats were randomly divided into normal group(n=6)and model group(n=54).30 successful rats were randomly divided into model group(MG),Atorvastatin group(AC),Shuangqing Pinghua Formula high dose group(SQPHF-H),Shuangqing Pinghua Formula middle dose group(SQPHF-M)and Shuangqing Pinghua Formula low dose group(SQPHF-L),with 6 rats in each group.The rats in each group were fed with corresponding feed(normal group normal diet,other groups high fat diet),the drugs were intervened for 8 weeks,and the drinking water of each group during the medication period was the same as that during the modeling period.During the experiment,the general condition of rats in each group was observed before and after administration,the weight of rats was weighed,the abdominal circumference and body length of rats were measured and the Lee's index of rats was calculated.At the end of the experiment,the body weight and liver wet weight of the rats were weighed,the liver index was calculated,the blood of the abdominal aorta was collected by collecting blood vessels without anticoagulants,and the serum was collected by centrifugation.Adiponectin(ADPN),free fatty acid(FFA),fasting insulin(FINS),fasting plasma glucose(FPG),total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)were measured,and insulin resistance index(HOMA-IR)was calculated.The liver tissue was fixed with 4% paraformaldehyde and 20% sucrose water respectively,and the pathological sections of liver tissue were made,and the morphological changes were observed.SPSS25.0 statistical software was used to analyze.According to the analysis results,the effects of Shuangqing Pinghua Formula on abdominal obesity,blood lipids and blood glucose and the improvement of insulin resistance(IR)in MS rats were evaluated.Results 1 compared with the normal group,the body hair of the model group gradually became yellow,less glossy,occasionally depilated,diet and water intake increased to a certain extent,urine volume increased,stool,lazy,slow response;compared with the model group,the above conditions of rats in Shuangqing Pinghua Formula group were improved gradually.2 Compared with the normal group,the body weight,abdominal circumference and Lee's index of the model group increased(P<0.05),while the body weight,abdominal circumference and Lee's index of the rats in the middle and high dose groups of Shuangqing Pinghua Formula decreased compared with the model group(P<0.05).3 Compared with the normal group,the levels of FPG,FINS and HOMA-IR in the model group increased(P<0.05),Compared with the model group,the level of FPG in the middle and high dose groups of Shuangqing Pinghua Formula decreased,and the FINS and HOMA-IR in the high dose group decreased(P<0.05).4 compared with the normal group,the levels of serum FFA,LDLC,TC,TG and liver index in the model group increased,while the levels of HDL-C and ADPN decreased(P<0.05).Compared with the model group,the levels of serum FFA,LDLC,TC,TG and liver index in Shuangqing Pinghua Formula group decreased,while the levels of HDL-C and ADPN increased(P<0.05).5 Compared with the normal group,the hepatocytes in the model group were steatosis,the degenerated hepatocytes were swollen in a round shape,and lipid droplet vacuoles could be seen in the cytoplasm.Compared with the model group,the arrangement of hepatocytes in each dose group of Shuangqing Pinghua Formula was more uniform and orderly,the vacuoles of lipid droplets in the cytoplasm decreased,and the steatosis and swelling of cells were improved.Conclusions 1 Shuangqing Pinghua Formula can effectively improve the central obesity and glucose and lipid metabolism of MS rats.2 Shuangqing Pinghua Formula can effectively improve the IR of MS rats.3 Shuangqing Pinghua Formula can decrease the level of FFA and increase the level of ADPN in MS rats,indicating that the mechanism of Shuangqing Pinghua Formula in regulating blood lipids in MS rats may be related to increasing the content of ADPN,increasing the oxidation of fatty acid and decreasing the level of FFA.Figure2;Table10;Reference 159...
