| Objectives Through the intervention of Shuangqing Pinghua Decoction on rats with metabolic syndrome model,observe its effect on blood pressure,serum NO,ET-1,AngⅡ,and insulin resistance,clarify its antihypertensive effect,explore the molecular mechanism of the prescription for reducing blood pressure,and provide experimental basis for the clinical treatment of metabolic syndrome.Methods 1 60 male SD rats with 4 weeks old and weighing 140-160 g,6 rats were randomly selected as the normal group,fed with normal feed and pure water,and the other54 were fed with high-fat feed+10% fructose water.The L-NAME with 20mg/kg/d was given at 11 th week of the experiment.Gavage for 2 weeks.At the 13 th week of the experiment,the STZ with 25mg/kg was injected intraperitoneally to induce the MS model.By the end of the 14 th week of modeling,the model was evaluated.2 At the end of modeling,MS rats meeting the modeling criteria were randomly divided into model group,valsartan group,Shuangqing Pinghua Decoction with high,middle and low dose groups with 6 rats in each group.Each treatment group was given valsartan(7.2mg/kg/d)and high-(13.38g/kg/d),medium-(6.93g/kg/d),and low-dose(3.47g/kg/d)medicated feeds of Shuangqing Pinghua Decoction.Rats were sacrificed after 8 weeks of administration,and the changes in basic indicators such as blood pressure were evaluated during treatment.Serum was used to detect FBG,FINS,and NO,ET-1,AngⅡ to observe changes in blood indexes;HE and Masson staining was used to observe pathological changes of the aorta;Western Blot was used to detect kidney tissue ACE,AT1 R,AT2R.SPSS 26.0 statistical software was used for statistical analysis of the data.Results 1 At the 14 th week of modeling,the blood pressure,weight,abdominal circumference,Lee’s index and blood glucose of the model were significantly higher than those of the normal group,and the difference was statistically significant(P <0.05).2 After8 weeks of drug intervention,the results showed that: in terms of obesity-related indicators,compared with the model group,there was statistical significance in the weight,abdominal circumference,Lee’s index of Shuangqing Pinghua Decoction high-,middle-dose group(P<0.05);in terms of blood pressure,compared with the model group,SBP and DBP in the high-dose group of Shuangqing Pinghua Decoction were significantly reduced,the difference was statistically significant(P<0.05),and the blood pressure level of each Chinese medicine group was inversely proportional to the dose;in terms of glucose metabolism,compared with the model group,there was statistical significance that the FBG,FINS and HOMA-IR in the high-dose group of Shuangqing Pinghua Decoction were reduced(P<0.05),and the levels of FBG,FINS and HOMA-IR in each Chinese medicine group were inversely proportional to the dose;in terms of vascular endothelial function,compared with the model group,the content of NO was increased and the content of ET-1and AngⅡ were reduced in Shuangqing Pinghua Decoction high-dose group,the difference was statistically significant(P<0.05),and each the dose group showed a doseeffect relationship.3 HE staining microscope showed that compared with the normal group,the aortic intima and media in the model group were significantly thickened to form plaques,and some of the cell swelling structures were ruptured,showing scattered cholesterol crystals;compared with the model group,the morphological structure of endothelial cells and smooth muscle cells,as well as the pathological changes of vascular intima and media in each treatment group were significantly improved,with the SPHD-Hgroup and the VC group being the most significant.Masson staining microscope showed that compared with the normal group,the model group had obvious collagen deposition in the whole layer of the vascular wall,the blood vessel cells and elastic fibers were arranged disorderly,and the tube wall thickened;compared with the model group,collagen deposition in thoracic aorta was improved in all treatment groups,with the SPHD-H group and the VC group having the most significant improvement.4 Western Blot method results showed that: compared with the normal group,the expression of ACE and AT1 R protein in the kidney tissue of the model group were significantly increased,and the expression of AT2 R protein was significantly reduced;compared with the model group,the ACE and AT1 R protein expression levels in kidney tissue of Shuangqing Pinghua Decoction group and valsartan group were significantly reduced,and AT2 R protein expression level was significantly increased.Meanwhile,the protein expression levels of ACE,AT1 R,and AT2 R in kidney tissues of Shuangqing Pinghua Decoction high-dose group were similar to valsartan.Conclusions 1 Shuangqing Pinghua Decoction can reduce blood pressure and blood glucose in MS rats,improve central obesity and insulin resistance.2 Shuangqing Pinghua Decoction can restore the balance of ET-1/NO,improve the function of vascular endothelium,and have a certain protective effect on blood vessels.3 Shuangqing Pinghua Decoction can down-regulate the expression of kidney ACE and AT1 R protein,upregulate the expression of kidney AT2 R protein,reduce the level of serum AngⅡ,and speculate that its antihypertensive mechanism may be related to the
