Objective: To observe the relationship between high-fat diet-induced renal injury and inflammatory reaction in rabbits and the intervention effect of Allisartan.Methods:After one week adaptive feed,15 healthy male New Zealand rabbits were randomly divided into 3 groups: Normal control group(NC)was fed with general diet;hyperlipidic diet group(HLD)were fed with high-fat diet(The experimental diet was composed of 1% cholesterol,6% peanut oil and 93% normal diet);Allisartan intervention group(ALST)were fed with high-fat diet and Allisartan 40mg/kg.d;NC and HLD were given the same amount of NS.The animals were sacrificed after 13-week feeding.The daily food intake and body weight of the three groups were compared.The blood lipids(total cholesterol TC,triglyceride TG,high-density lipoprotein HDL and low-density lipoprotein LDL),body weight and renal function(creatinine Cr,blood urea nitrogen,BUN)were measured in each group.ELISA was adopted to test the interleukin-6(IL-6)and interleukin-1 beta(IL-1?)in serum.One side of the kidney was selected to make paraffin Sections,HE staining,PAS staining and Masson staining were used to study the renal pathological changes;immunohistochemistry was used to detect the expression of ?-smooth muscle actin(?-SMA)and Desmin in the kidney.After statistic SPSS19.0 software analyzing,obtained relative conclusions.The comparison among groups was performed by simple factor analysis of variance,P<0.05 had statistical significance.Results:1.There was no significant difference in the food intake of the three groups(P> 0.05).The body weight,TC,TG,LDL-C,Scr and BUN levels in HLD group and ALST group were significantly higher and HDL-C lower than those in NC group.Compared with the HLD group,the level of Scr and BUN were significantly lower than that of ALST Group(P<0.05);while the level of TC,TG,LDL-C,HDL-C were showed no significant difference in HLD group and ALST Group(P>0.05).2.HE staining the results showed that Allisartan can improve the morphological changes of kidney tissue induced by high-lipid diet,improve mesangial proliferation and glomerulor sclerosis,glycogen staining showed decreased glomerular glycogen deposition and decreased glomerular erythema.Masson staining showed decreased blue-stained collagen fiber of renal tubular.3.Immunohistologic staining of ?-SMA and Desmin showed that the positive expression of HLD and ALST group was stronger than that of NC group(P<0.05),and the positive expression was weaker in ALST group than HLD group(P<0.05).4.The levels of IL-6 and IL-1? in HLD and ALST group were significantly higher than those in NC Group(P<0.05).The levels of IL-6 and IL-1? in ALST group were significantly lower than those in HLD Group(P<0.05).Conclusion:1.The results of this study showed that Hyperlipidemia can induce some cell protein of rabbit kidney expression abnormally and damage the kidney.2.Allisartan can alleviate the chronic kidney injury caused by high-fat diet,and the mechanism relate to its effect on inflammatory reaction.
