Study On Alkaloids And Antioxidant Activity Of Consolida Ajacis(L.)

Consolida ajacis(L.)Schur belongs to the genus Consolida,which is also kn own as the delphinium,and so on.As one traditonal chinese medicine,the roots of Consolida ajacis(L.)Schur have variety bioactivities,such as analgesia,antiba cterial,anti-inflammatory and anti-tumor.In this paper,the chemical constituents from the roots of Consolida ajacis(L.)Schur were systematic investigated,the ex traction conditions of total alkaloids were optimized by response surface methodol ogy,and the vitro antioxidant activity of the total alkaloids were also studied.The whole plant was extracted by ethanol diacolation,and the total extract was obtained by rotary evaporation.The total alkaloid extract of Consolida ajacis(L.)Schur was obtained by acid-base extraction.Four fractions(Fr.1-4)were obtained by silica gel column chromatography eluting with a gradient of chlorofor m-methanol.Five compounds(FYC-1,FYC-2,FYC-3,FYC-4,FYC-5)were obtain ed from Fr.2 which was purified by repeated column chromatography such as M CI,ODS,Sephadex LH-20 and PHPLC.The structures were elucidated by UV,IR,MS,XRD,1D and 2D NMR.FYC-1,FYC-2,FYC-3,FYC-4,FYC-5 were identif ied as lycoctonine,4-(2-(methoxycarbonyl)phenyl)amino)-2-methyl-4-oxobutanoic aci d,4-((2-(methoxycarbonyl)phenyl)amino)-3-methyl-4-oxobutanoic acid,anthroylylcoct one,3?,23-Dihydroxyursa-12,18(19)-dien-28-oic acid 28-?-D-glucopyranosyl ester.Ultrasound-assisted ethanol solvent method was used to extract the total alkal oids of Consolida ajacis(L.)Schur.The optimum extraction conditions were deter mined by single factor experiment and response surface method: the ratio of solve nt to sample was 21:1,the extraction time was 52 min,and the ethanol volume f raction was 72%,p H value of 2.0,extraction temperature of 60°C.Under this con dition,the extraction rate of total alkaloids of Consolida ajacis(L.)Schur was 0.014%.The study in vitro antioxidant activity of the total alkaloids of Consolida ajacis(L.)Schur was evaluated.The results showed that the total alkaloids of Consolida ajacis(L.)Schur had good scavenging ability to DPPH free radicals,while the effect of scavenginghydroxyl radicals and superoxide anion is not obvious.
