Zhitong Huazheng capsules(ZTHZC)are made from 19 kinds of Chinese medicines,such as Codonopsis radix.Clinically,it mainly treats gynecological diseases such as irregular menstruation,dysmenorrhea and chronic pelvic inflammatory disease,with few adverse reactions and significant curative effects.At present,the effective ingredients of the drug are unclear and the mechanism of action is unclear.Therefore,it is necessary to carry out its secondary development.This article is mainly carried out from two aspects.On the one hand,it further improves the material basis of the anti-blood stasis of Zhitong Huazheng capsules;on the one hand,it studies the mechanism of anti-blood stasis of Zhitong Huazheng capsules and related gynecological diseases.The clinical application of Zhitong Huazheng capsule provides more basis and reference,and also lays the foundation for the further development of Zhitong Huazheng capsule.(1)Based on UPLC-Q-TOF-MS technology and UNIFI natural product analysis platform to analyze the chemical composition of Zhitong Huazheng capsule.For the first time,UPLC-Q-TOF-MS technology combined with the UNIFI natural product analysis platform was used to analyze 80% of the methanol extracts of Zhitong Huazheng capsules.A total of 70 compounds were identified,including organic acids,flavones,and alkaloids.(2)Based on UPLC-Q-TOF / MS technology to analyze the serum transition components of Zhitong Huazheng capsules.UPLC-Q-TOF-MS technology combined with multivariate statistical analysis was used for the first time to study the serum transition components of Zhitong Huazheng Capsules.15 serum transition components were identified,all of which were prototype components,including organic acids,flavones,and triterpenoid saponins.,Alkaloids and other substances.They are Ononin,Danshensu,Formononetin,salvianolic acids A,B,C,D,9,12-dihydroxy-15-nineteen carbonate,luteolin,palm Ethyl oleate,linoleic acid,tetrahydropalmatine,corydaline,astragaloside IV and astragaloside?,these components may be closely related to the role of Zhitong Huazheng capsules in treating gynecological diseases.(3)The anti-blood stasis activity of Zhitong Huazheng Capsule was evaluated by spectrum-effect relationship method,and its anti-blood stasis active ingredients were screened.In this study,HPLC fingerprints of extracts with different solvent ratios of Zhitong Huazheng Capsules were constructed.32 chromatographic peaks were identified as common peaks,and their similarities were evaluated.The similarities were greater than 0.90.The spectra-effect relationship were evaluated by gray correlation analysis(GRA).8 anti-blood stasis active ingredients were screened out,and their contribution rates were all greater than 0.80,and they had significant contributions in anti-blood stasis.Study on the endogenous substances related to the anti-blood stasis activity of Zhitong Huazheng Capsule based on metabolomics.(4)Research on the related endogenous substances of Zhitong Huazheng Capsule against blood stasis based on metabolomics.Using UPLC-Q / TOF-MS technology and multivariate statistical analysis for the first time to explore the mechanism of anti-blood stasis activity of Zhitong Huazheng capsules.The results indicate that 25 differential metabolites identified from plasma,urine,and feces may be closely related to the BSS model.Differential metabolites are involved in 7 metabolic pathways of arachidonic acid,?-linolenic acid,retinol,glycerophospholipids,steroid hormone synthesis,sphingolipids and linoleic acid.The above results help to understand the pathogenesis of blood stasis syndrome,find the target of clinical diagnosis,clarify the action mechanism of ZTHZC,and provide further evidence for using ZTHZC as an anti-blood stasis drug.Based on the chemical composition of the whole composition,blood composition analysis and spectrum-effect relationship screening,based on the network pharmacology,the potential active ingredients and action mechanism of the Zhitong Huazheng Capsule for the treatment of gynecological related diseases were screened.(5)Based on the full ingredients that complies with the five principles of generic drugs,blood ingredients and ingredients screened for spectral effect relationships.Screening the potential active ingredients and mechanism of Zhitong Huazheng capsules for treating gynecology-related diseases based on network pharmacology.Using network pharmacology technology for the first time to explore the mechanism of action of Zhitong Huazheng capsules on gynecological diseases such as dysmenorrhea,irregular menstruation and pelvic inflammatory disease.The results show that 39 active ingredients are involved in 179 targets,and 7 key active ingredients of luteolin,Wogonin,tetrahydropalmatine,corydaline,kaempferol,linoleic acid and ethyl p-methoxycinnamic acid are predicted.And 8 key proteins such as PTGS2,PPARG,MMP9,CASP3,TNF,RELA,JUN,AR,etc..Zhitong Huazheng Capsules may treat diseases such as dysmenorrhea,irregular menstruation and pelvic inflammatory disease by regulating multiple pathways including Pathways in cancer,PI3K-Akt signaling pathway,and TNF signaling pathway,and improve the patient's condition.In summary,this thesis has thoroughly studied the composition of Zhitong Huazheng capsules,filling in the research of blood-injection components of Zhitong Huazheng capsules,and used the spectrum-effect relationship for the first time to screen out anti-blood stasis active ingredients,UPLC-Q/TOF-MS technology combined with multivariate statistical analysis to explore the mechanism of anti-blood stasis activity of Zhitong Huazheng Capsule Explore the molecular mechanism and main active ingredients of Zhitong Huazheng Capsules for treating gynecological diseases such as dysmenorrhea,endometriosis and pelvic inflammation based on network pharmacology technology.The thesis systematically explores the basic research of pharmacodynamic substance of Zhitong Huazheng capsules and discusses its mechanism of action,which provides a reference and scientific basis for further development and clinical application.
