Lichen sclerosus et atrophicus(LSA)is a chronic inflammatory skin disease of unknown etiology,with a potential for atrophy,destructive scarring,functional impairment,and malignantevolution.LSA is a dynamically changing skin disease,which has a variety of histological manifestations.Due to the lack of pathological diagnostic techniques or experience of clinicians,LSA is often misdiagnosed as sclerosing and atrophic morphea.Therefore,it is urgent to develop a new technique accurate identification of LSA skin diseases.In this paper,we applied multiphoton microscope(MPM)to identify the two similar skin disease compared with the traditional histology method.Our results proved that MPM technique can be used to distinguish skin diseases,which will greatly promote the pathogenesis of sclerosing atrophic moss and scleroderma,as well as clinical diagnosis and treatment of related research.Firstly,MPM was used to visualize the human skin epidermal microstructure,including elastin,collagen in the dermis,all of which are closely related to human skin diseases.Secondly,we obtained histopathological characteristics of LSA images with high resolution and contrast,including hyperkeratotic epidermis,homogenous collagen fibers,and infiltrating inflammatory cells.Finally,MPM was used to visualize the microstructures of LSA and morphea,and to quantify the content of extracellular matrix and the direction of collagen fibers.This study shows that MPM can not only clearly show microstructure and morphological changes in skin tissue,but also provide additional pathologic information in the diagnosis of two kinds of skin diseases.As a newly developed technology,MPM is being widely recognized and applied.The non-invasive inspection features of MPM will have broad application prospects in dermatology.With the development and improvement of this technology,we have good reason to believe that MPM will promote the etiology and pathogenesis of skin diseases,provide effective methods for diagnosis and treatment,as well as promote the development of dermatology.
