Objective: To study the anti apoptotic molecular mechanism of Astragalus Polysaccharides(APS)on the injury of rat cardiomyocytes(H9c2)induced by uremic toxin indole sulfate,and to observe its effect on the expression of Bax,Bcl-2,Caspase-3,Caspase-9 and other major regulatory factors in the mitochondrial apoptotic pathway,so as to provide theoretical basis for the clinical application of APS in cardiovascular injury caused by chronic kidney disease.Methods: H9c2 cells were cultured in vitro.The experiment was divided into blank control group,indoxyl sulfate group,indoxyl sulfate + astragalus polysaccharides group.Cell morphology was observed under inverted microscope,cell viability was detected by CCK-8,ROS level was detected by fluorescence spectrophotometer,apoptosis rate was detected by TUNEL staining,mRNA expression of Bax,Bcl-2,Caspase-3 and Caspase-9 were detected by RT-qPCR,Bax,Bcl-2,caspase-3 and caspase-9 were detected by Western Blot Protein expression level.Results: Compared with the blank control group,indoxyl sulfate group could significantly reduce the cell viability of H9c2 and increase the content of ROS in cells(P<0.01).Compared with indoxyl sulfate group,in the indoxyl sulfate + astragalus polysaccharides group with different concentrations,as the concentration of the astragalus polysaccharides increased,the cell viability gradually increased,and the ROS content gradually decreased(P<0.01).In addition,compared with the indoxyl sulfate group,the indoxyl sulfate + astragalus polysaccharide group with different concentrations could significantly reduce the mRNA and protein expression levels of Bax?Caspase-3 and Caspase-9(P<0.01),and increased Bcl-2 level(P<0.01).Conclusion: Indoxyl sulfate may induce H9c2 cell damage by oxidative stress,while astragalus polysaccharides may improve H9c2 cell damage by reducing ROS level and regulating Bax,Bcl-2,Caspase-3,Caspase-9 gene and protein expression level in cells.It is suggested that the mechanism of Astragalus Polysaccharides on uremic toxin induced cardiomyocyte injury is based on mitochondrial apoptosis pathway,which can promote the survival of damaged cardiomyocytes through anti apoptosis,so as to reduce the cardiovascular damage caused by chronic kidney disease.
