Objective: Study the effects and mechanism of kunlun snow chrysanthemum water extraction liquid on lipid oxidative damage of liver in hypoxia and phlegm turbidity syndrome experimental rat.Methods: Forty SPF male SD rats were randomly divided into hypoxia co ntrol(H)group,hypoxia and high fat diet(HHFD)group,kunlun snow chrysa nthemum intervention H(XJH)group and kunlun snow chrysanthemum interve ntion HHFD(XJHHFD)group.Rats of four groups were placed in a hypobaric hypoxia chamber simulating an environment at an altitude of 5000 m.At 8:00 every day,they were given relevant dietary intervention(intragastric dose 10ml/kg);at 8:00 pm,they were given intragastric intervention with normal sali ne or Kunlun snow chrysanthemum water extraction(intragastric dose 10 ml/k g).After 1 week of the experiment,the levels of superoxide dismutase(SOD),malondialdehyde(MDA),nitric oxide lyase(NOS)m RNA,lipid parameters,and nitric oxide(NO)in liver tissue were measured,and the pathomorphological c hanges of liver tissue were observed microscopically.Results: Compared with the H group,TCH,LDL,MDA,NO,and i NOS m RNA were significantly increased(P<0.01),while e NOS m RNA was decreas ed(P<0.01)in the HHFD group;e NOS m RNA was decreased(P<0.01)in the XJH group.Compared with the HHFD group,MDA,i NOS m RNA and NO were decreased(P<0.01),SOD and n NOS m RNA were increased(P<0.01)in t he XJHHFD group.Compared with the XJH group,MDA decreased(P<0.01),NO and n NOS m RNA increased(P<0.01 or P<0.05)in the XJHHFD group.Conclusion: Hypoxia has a synergistic effect with hypoxia,which can ag gravate abnormal lipid metabolism and liver tissue injury;the change of SOD/MDA level proves that the damage of hypoxia combined with diet on liver tis sue of experimental rats is related to oxidative stress;the detection of NOS/N O level reveals that the oxidative stress effect of hypoxia or diet is closely rel ated to the increase of i NOS;The aqueous extract of kunlun snow chrysanthe mum had a significant protective effect on oxidative stress injury in the liver o f rats fed with hypoxia combined with diet.
