Photodynamic therapy is a non-invasive cancer treatment using photosensitizers and laser.However,many photosensitizers are hydrophobic drugs and easily aggregate in aqueous solution.This not only reduces the efficiency of photodynamic therapy,but also results in a short blood circulation half-life of the photosensitizer molecules,so they cannot be delivered to tumor tissue and metabolized out of the body.Therefore,choosing a suitable drug carrier to improve the bioavailability of the photosensitizer is of clinical application value.Based on the problems,this work prepare a zeolite imidazolate framework-8(ZIF-8)nanocomposite modified with hyaluronic acid(HA)by one-pot encapsulation and self-assembly to deliver Ce6(ZIF-8@Ce6-HA)for photodynamic therapy.ZIF-8@Ce6-HA exhibits acceptable encapsulation efficiency,effective cell uptake and good biocompatibility.In addition,ZIF-8 solves the problem that Ce6 molecules are easy to reunite in the physiological environment,thereby improving the efficiency of photodynamic therapy.Antitumor experiments in vitro showed that the ZIF-8@Ce6-HA group had higher cytotoxicity than the Ce6 group after laser irradiation,and most of cancer cells were killed by reactive oxygen species generated during photodynamic therapy.In addition,inductively coupled plasma mass spectrometry and toxicity tests in vitro and in vivo showed that modification of HA increased blood circulation time and reduced systemic toxicity of ZIF-8.In summary,this work has developed a nano-delivery platform for photodynamic therapy with potential for clinical application.
